age A differentiable simulation package for performing inference of synchrotron-radiation-based diagnostics By journals.iucr.org Published On :: 2024-02-16 The direction of particle accelerator development is ever-increasing beam quality, currents and repetition rates. This poses a challenge to traditional diagnostics that directly intercept the beam due to the mutual destruction of both the beam and the diagnostic. An alternative approach is to infer beam parameters non-invasively from the synchrotron radiation emitted in bending magnets. However, inferring the beam distribution from a measured radiation pattern is a complex and computationally expensive task. To address this challenge we present SYRIPY (SYnchrotron Radiation In PYthon), a software package intended as a tool for performing inference of synchrotron-radiation-based diagnostics. SYRIPY has been developed using PyTorch, which makes it both differentiable and able to leverage the high performance of GPUs, two vital characteristics for performing statistical inference. The package consists of three modules: a particle tracker, Lienard–Wiechert solver and Fourier optics propagator, allowing start-to-end simulation of synchrotron radiation detection to be carried out. SYRIPY has been benchmarked against SRW, the prevalent numerical package in the field, showing good agreement and up to a 50× speed improvement. Finally, we have demonstrated how SYRIPY can be used to perform Bayesian inference of beam parameters using stochastic variational inference. Full Article text
age Synchrotron infrared nanospectroscopy in fourth-generation storage rings By journals.iucr.org Published On :: 2024-04-17 Fourth-generation synchrotron storage rings represent a significant milestone in synchrotron technology, offering outstandingly bright and tightly focused X-ray beams for a wide range of scientific applications. However, due to their inherently tight magnetic lattices, these storage rings have posed critical challenges for accessing lower-energy radiation, such as infrared (IR) and THz. Here the first-ever IR beamline to be installed and to operate at a fourth-generation synchrotron storage ring is introduced. This work encompasses several notable advancements, including a thorough examination of the new IR source at Sirius, a detailed description of the radiation extraction scheme, and the successful validation of our optical concept through both measurements and simulations. This optimal optical setup has enabled us to achieve an exceptionally wide frequency range for our nanospectroscopy experiments. Through the utilization of synchrotron IR nanospectroscopy on biological and hard matter samples, the practicality and effectiveness of this beamline has been successfully demonstrated. The advantages of fourth-generation synchrotron IR sources, which can now operate with unparalleled stability as a result of the stringent requirements for producing low-emittance X-rays, are emphasized. age The effect of transport apertures on relay-imaged, sharp-edged laser profiles in photoinjectors and the impact on electron beam properties By journals.iucr.org Published On :: 2024-06-06 In a photoinjector electron source, the initial transverse electron bunch properties are determined by the spatial properties of the laser beam on the photocathode. Spatial shaping of the laser is commonly achieved by relay imaging an illuminated circular mask onto the photocathode. However, the Gibbs phenomenon shows that recreating the sharp edge and discontinuity of the cut profile at the mask on the cathode is not possible with an optical relay of finite aperture. Furthermore, the practical injection of the laser into the photoinjector results in the beam passing through small or asymmetrically positioned apertures. This work uses wavefront propagation to show how the transport apertures cause ripple structures to appear in the transverse laser profile even when effectively the full laser power is transmitted. The impact of these structures on the propagated electron bunch has also been studied with electron bunches of high and low charge density. With high charge density, the ripples in the initial charge distribution rapidly wash-out through space charge effects. However, for bunches with low charge density, the ripples can persist through the bunch transport. Although statistical properties of the electron bunch in the cases studied are not greatly affected, there is the potential for the distorted electron bunch to negatively impact machine performance. age Signal-to-noise and spatial resolution in in-line imaging. 1. Basic theory, numerical simulations and planar experimental images By journals.iucr.org Published On :: 2024-06-06 Signal-to-noise ratio and spatial resolution are quantitatively analysed in the context of in-line (propagation based) X-ray phase-contrast imaging. It is known that free-space propagation of a coherent X-ray beam from the imaged object to the detector plane, followed by phase retrieval in accordance with Paganin's method, can increase the signal-to-noise in the resultant images without deteriorating the spatial resolution. This results in violation of the noise-resolution uncertainty principle and demonstrates `unreasonable' effectiveness of the method. On the other hand, when the process of free-space propagation is performed in software, using the detected intensity distribution in the object plane, it cannot reproduce the same effectiveness, due to the amplification of photon shot noise. Here, it is shown that the performance of Paganin's method is determined by just two dimensionless parameters: the Fresnel number and the ratio of the real decrement to the imaginary part of the refractive index of the imaged object. The relevant theoretical analysis is performed first, followed by computer simulations and then by a brief test using experimental images collected at a synchrotron beamline. Finding an appropriate ηc demands too much computing time because MOGA needs be run several times in order to find a good ηc. In this paper, a self-adaptive crossover parameter is introduced in a strategy to adopt a new ηc for every generation while running MOGA. This new scheme has also been adopted for a multi-generation Gaussian process optimization (MGGPO) when producing trial solutions. Compared with the existing MGGPO and MOGA, the MGGPO and MOGA with the new strategy show better performance in nonlinear optimization for the design of low-emittance storage rings. Full Article text

The feasibility of retrieving the surface error of a lens system by using only the intensity of the propagated beam at several distances is demonstrated. A neural network, trained with a few thousand simulations using random errors, can predict accurately the lens error profile that accounts for all aberrations. age New opportunities for time-resolved imaging using diffraction-limited storage rings By journals.iucr.org Published On :: 2024-07-30 The advent of diffraction-limited storage rings (DLSRs) has boosted the brilliance or coherent flux by one to two orders of magnitude with respect to the previous generation. One consequence of this brilliance enhancement is an increase in the flux density or number of photons per unit of area and time, which opens new possibilities for the spatiotemporal resolution of X-ray imaging techniques. This paper studies the time-resolved microscopy capabilities of such facilities by benchmarking the ForMAX beamline at the MAX IV storage ring. It is demonstrated that this enhanced flux density using a single harmonic of the source allows micrometre-resolution time-resolved imaging at 2000 tomograms per second and 1.1 MHz 2D acquisition rates using the full dynamic range of the detector system. X-ray speckle-based techniques stand out among other in situ techniques for their easy experimental setup and various data acquisition modes. Although X-ray speckle-based techniques have been under development for more than a decade, there are still no user-friendly software packages for new researchers to begin with. Here, we present an open-source Python package, spexwavepy, for X-ray wavefront sensing using speckle-based techniques. This Python package covers a variety of X-ray speckle-based techniques, provides plenty of examples with real experimental data and offers detailed online documentation for users. age Characterizing electron-collecting CdTe for use in a 77 ns burst-rate imager By journals.iucr.org Published On :: 2024-08-07 The Keck-PAD (pixel array detector) was developed at Cornell as a burst-rate imager capable of recording images from successive electron bunches (153 ns period) from the Advanced Photon Source (APS). Both Si and hole-collecting Schottky CdTe have been successfully bonded to this ASIC (application-specific integrated circuit) and used with this frame rate. The facility upgrades at the APS will lower the bunch period to 77 ns, which will require modifications to the Keck-PAD electronics to image properly at this reduced period. In addition, operation at high X-ray energies will require a different sensor material having a shorter charge collection time. For the target energy of 40 keV for this project, simulations have shown that electron-collecting CdTe should allow >90% charge collection within 35 ns. This collection time will be sufficient to sample the signal from one frame and prepare for the next. 750 µm-thick electron-collecting Schottky CdTe has been obtained from Acrorad and bonded to two different charge-integrating ASICs developed at Cornell, the Keck-PAD and the CU-APS-PAD. Carrier mobility has been investigated using the detector response to single X-ray bunches at the Cornell High Energy Synchrotron Source and to a pulsed optical laser. The tests indicate that the collection time will meet the requirements for 77 ns imaging. Full Article text
age Comparing single-shot damage thresholds of boron carbide and silicon at the European XFEL By journals.iucr.org Published On :: 2024-08-25 Xray free-electron lasers (XFELs) enable experiments that would have been impractical or impossible at conventional X-ray laser facilities. Indeed, more XFEL facilities are being built and planned, with their aim to deliver larger pulse energies and higher peak brilliance. While seeking to increase the pulse power, it is quintessential to consider the maximum pulse fluence that a grazing-incidence FEL mirror can withstand. To address this issue, several studies were conducted on grazing-incidence damage by soft X-ray FEL pulses at the European XFEL facility. Boron carbide (B4C) coatings on polished silicon substrate were investigated using 1 keV photon energy, similar to the X-ray mirrors currently installed at the soft X-ray beamlines (SASE3). age HEIDI: an experiment-management platform enabling high-throughput fragment and compound screening By journals.iucr.org Published On :: 2024-04-12 The Swiss Light Source facilitates fragment-based drug-discovery campaigns for academic and industrial users through the Fast Fragment and Compound Screening (FFCS) software suite. This framework is further enriched by the option to utilize the Smart Digital User (SDU) software for automated data collection across the PXI, PXII and PXIII beamlines. In this work, the newly developed HEIDI webpage (https://heidi.psi.ch) is introduced: a platform crafted using state-of-the-art software architecture and web technologies for sample management of rotational data experiments. The HEIDI webpage features a data-review tab for enhanced result visualization and provides programmatic access through a representational state transfer application programming interface (REST API). The migration of the local FFCS MongoDB instance to the cloud is highlighted and detailed. This transition ensures secure, encrypted and consistently accessible data through a robust and reliable REST API tailored for the FFCS software suite. Collectively, these advancements not only significantly elevate the user experience, but also pave the way for future expansions and improvements in the capabilities of the system. Full Article text
age STOPGAP: an open-source package for template matching, subtomogram alignment and classification By journals.iucr.org Published On :: 2024-04-12 Cryo-electron tomography (cryo-ET) enables molecular-resolution 3D imaging of complex biological specimens such as viral particles, cellular sections and, in some cases, whole cells. This enables the structural characterization of molecules in their near-native environments, without the need for purification or separation, thereby preserving biological information such as conformational states and spatial relationships between different molecular species. Subtomogram averaging is an image-processing workflow that allows users to leverage cryo-ET data to identify and localize target molecules, determine high-resolution structures of repeating molecular species and classify different conformational states. Here, STOPGAP, an open-source package for subtomogram averaging that is designed to provide users with fine control over each of these steps, is described. In providing detailed descriptions of the image-processing algorithms that STOPGAP uses, this manuscript is also intended to serve as a technical resource to users as well as for further community-driven software development. age Managing macromolecular crystallographic data with a laboratory information management system By journals.iucr.org Published On :: 2024-07-10 Protein crystallography is an established method to study the atomic structures of macromolecules and their complexes. A prerequisite for successful structure determination is diffraction-quality crystals, which may require extensive optimization of both the protein and the conditions, and hence projects can stretch over an extended period, with multiple users being involved. The workflow from crystallization and crystal treatment to deposition and publication is well defined, and therefore an electronic laboratory information management system (LIMS) is well suited to management of the data. Completion of the project requires key information on all the steps being available and this information should also be made available according to the FAIR principles. As crystallized samples are typically shipped between facilities, a key feature to be captured in the LIMS is the exchange of metadata between the crystallization facility of the home laboratory and, for example, synchrotron facilities. On completion, structures are deposited in the Protein Data Bank (PDB) and the LIMS can include the PDB code in its database, completing the chain of custody from crystallization to structure deposition and publication. A LIMS designed for macromolecular crystallography, IceBear, is available as a standalone installation and as a hosted service, and the implementation of key features for the capture of metadata in IceBear is discussed as an example. age Surface-mutagenesis strategies to enable structural biology crystallization platforms By journals.iucr.org Published On :: 2024-08-29 A key prerequisite for the successful application of protein crystallography in drug discovery is to establish a robust crystallization system for a new drug-target protein fast enough to deliver crystal structures when the first inhibitors have been identified in the hit-finding campaign or, at the latest, in the subsequent hit-to-lead process. The first crucial step towards generating well folded proteins with a high likelihood of crystallizing is the identification of suitable truncation variants of the target protein. In some cases an optimal length variant alone is not sufficient to support crystallization and additional surface mutations need to be introduced to obtain suitable crystals. In this contribution, four case studies are presented in which rationally designed surface modifications were key to establishing crystallization conditions for the target proteins (the protein kinases Aurora-C, IRAK4 and BUB1, and the KRAS–SOS1 complex). The design process which led to well diffracting crystals is described and the crystal packing is analysed to understand retrospectively how the specific surface mutations promoted successful crystallization. The presented design approaches are routinely used in our team to support the establishment of robust crystallization systems which enable structure-guided inhibitor optimization for hit-to-lead and lead-optimization projects in pharmaceutical research. age EMhub: a web platform for data management and on-the-fly processing in scientific facilities By journals.iucr.org Published On :: 2024-10-07 Most scientific facilities produce large amounts of heterogeneous data at a rapid pace. Managing users, instruments, reports and invoices presents additional challenges. To address these challenges, EMhub, a web platform designed to support the daily operations and record-keeping of a scientific facility, has been introduced. EMhub enables the easy management of user information, instruments, bookings and projects. The application was initially developed to meet the needs of a cryoEM facility, but its functionality and adaptability have proven to be broad enough to be extended to other data-generating centers. The expansion of EMHub is enabled by the modular nature of its core functionalities. The application allows external processes to be connected via a REST API, automating tasks such as folder creation, user and password generation, and the execution of real-time data-processing pipelines. EMhub has been used for several years at the Swedish National CryoEM Facility and has been installed in the CryoEM center at the Structural Biology Department at St. Jude Children's Research Hospital. A fully automated single-particle pipeline has been implemented for on-the-fly data processing and analysis. At St. Jude, the X-Ray Crystallography Center and the Single-Molecule Imaging Center have already expanded the platform to support their operational and data-management workflows. Full Article text
age The ABC toxin complex from Yersinia entomophaga can package three different cytotoxic components expressed from distinct genetic loci in an unfolded state: the structures of both shell and cargo By journals.iucr.org Published On :: 2024-05-01 Bacterial ABC toxin complexes (Tcs) comprise three core proteins: TcA, TcB and TcC. The TcA protein forms a pentameric assembly that attaches to the surface of target cells and penetrates the cell membrane. The TcB and TcC proteins assemble as a heterodimeric TcB–TcC subcomplex that makes a hollow shell. This TcB–TcC subcomplex self-cleaves and encapsulates within the shell a cytotoxic `cargo' encoded by the C-terminal region of the TcC protein. Here, we describe the structure of a previously uncharacterized TcC protein from Yersinia entomophaga, encoded by a gene at a distant genomic location from the genes encoding the rest of the toxin complex, in complex with the TcB protein. When encapsulated within the TcB–TcC shell, the C-terminal toxin adopts an unfolded and disordered state, with limited areas of local order stabilized by the chaperone-like inner surface of the shell. We also determined the structure of the toxin cargo alone and show that when not encapsulated within the shell, it adopts an ADP-ribosyltransferase fold most similar to the catalytic domain of the SpvB toxin from Salmonella typhimurium. Our structural analysis points to a likely mechanism whereby the toxin acts directly on actin, modifying it in a way that prevents normal polymerization. Full Article text
age Lattice response to the radiation damage of molecular crystals: radiation-induced versus thermal expansivity By journals.iucr.org Published On :: 2024-01-04 The interaction of intense synchrotron radiation with molecular crystals frequently modifies the crystal structure by breaking bonds, producing fragments and, hence, inducing disorder. Here, a second-rank tensor of radiation-induced lattice strain is proposed to characterize the structural susceptibility to radiation. Quantitative estimates are derived using a linear response approximation from experimental data collected on three materials Hg(NO3)2(PPh3)2, Hg(CN)2(PPh3)2 and BiPh3 [PPh3 = triphenylphosphine, P(C6H5)3; Ph = phenyl, C6H5], and are compared with the corresponding thermal expansivities. The associated eigenvalues and eigenvectors show that the two tensors are not the same and therefore probe truly different structural responses. The tensor of radiative expansion serves as a measure of the susceptibility of crystal structures to radiation damage. Full Article text

New treatments are needed because the currently used drug, ivermectin, is contraindicated in pregnant women and those co-infected with Loa loa. The Seattle Structural Genomics Center for Infectious Disease (SSGCID) produced, crystallized and determined the apo structure of N-terminally hexahistidine-tagged O. volvulus macrophage migration inhibitory factor-1 (His-OvMIF-1). OvMIF-1 is a possible drug target. His-OvMIF-1 has a unique jellyfish-like structure with a prototypical macrophage migration inhibitory factor (MIF) trimer as the `head' and a unique C-terminal `tail'. Deleting the N-terminal tag reveals an OvMIF-1 structure with a larger cavity than that observed in human MIF that can be targeted for drug repurposing and discovery. Removal of the tag will be necessary to determine the actual biological oligomer of OvMIF-1 because size-exclusion chomatographic analysis of His-OvMIF-1 suggests a monomer, while PISA analysis suggests a hexamer stabilized by the unique C-terminal tails. age Using XAS to monitor radiation damage in real time and post-analysis, and investigation of systematic errors of fluorescence XAS for Cu-bound amyloid-β By journals.iucr.org Published On :: 2024-02-01 X-ray absorption spectroscopy (XAS) is a promising technique for determining structural information from sensitive biological samples, but high-accuracy X-ray absorption fine structure (XAFS) requires corrections of systematic errors in experimental data. Low-temperature XAS and room-temperature X-ray absorption spectro-electrochemical (XAS-EC) measurements of N-truncated amyloid-β samples were collected and corrected for systematic effects such as dead time, detector efficiencies, monochromator glitches, self-absorption, radiation damage and noise at higher wavenumber (k). A new protocol was developed using extended X-ray absorption fine structure (EXAFS) data analysis for monitoring radiation damage in real time and post-analysis. The reliability of the structural determinations and consistency were validated using the XAS measurement experimental uncertainty. The correction of detector pixel efficiencies improved the fitting χ2 by 12%. An improvement of about 2.5% of the structural fitting was obtained after dead-time corrections. Normalization allowed the elimination of 90% of the monochromator glitches. The remaining glitches were manually removed. The dispersion of spectra due to self-absorption was corrected. Standard errors of experimental measurements were propagated from pointwise variance of the spectra after systematic corrections. Calculated uncertainties were used in structural refinements for obtaining precise and reliable values of structural parameters including atomic bond lengths and thermal parameters. age DLSIA: Deep Learning for Scientific Image Analysis By journals.iucr.org Published On :: 2024-03-21 DLSIA (Deep Learning for Scientific Image Analysis) is a Python-based machine learning library that empowers scientists and researchers across diverse scientific domains with a range of customizable convolutional neural network (CNN) architectures for a wide variety of tasks in image analysis to be used in downstream data processing. DLSIA features easy-to-use architectures, such as autoencoders, tunable U-Nets and parameter-lean mixed-scale dense networks (MSDNets). Additionally, this article introduces sparse mixed-scale networks (SMSNets), generated using random graphs, sparse connections and dilated convolutions connecting different length scales. For verification, several DLSIA-instantiated networks and training scripts are employed in multiple applications, including inpainting for X-ray scattering data using U-Nets and MSDNets, segmenting 3D fibers in X-ray tomographic reconstructions of concrete using an ensemble of SMSNets, and leveraging autoencoder latent spaces for data compression and clustering. As experimental data continue to grow in scale and complexity, DLSIA provides accessible CNN construction and abstracts CNN complexities, allowing scientists to tailor their machine learning approaches, accelerate discoveries, foster interdisciplinary collaboration and advance research in scientific image analysis. age Robust image descriptor for machine learning based data reduction in serial crystallography By journals.iucr.org Published On :: 2024-03-26 Serial crystallography experiments at synchrotron and X-ray free-electron laser (XFEL) sources are producing crystallographic data sets of ever-increasing volume. While these experiments have large data sets and high-frame-rate detectors (around 3520 frames per second), only a small percentage of the data are useful for downstream analysis. Thus, an efficient and real-time data classification pipeline is essential to differentiate reliably between useful and non-useful images, typically known as `hit' and `miss', respectively, and keep only hit images on disk for further analysis such as peak finding and indexing. While feature-point extraction is a key component of modern approaches to image classification, existing approaches require computationally expensive patch preprocessing to handle perspective distortion. This paper proposes a pipeline to categorize the data, consisting of a real-time feature extraction algorithm called modified and parallelized FAST (MP-FAST), an image descriptor and a machine learning classifier. For parallelizing the primary operations of the proposed pipeline, central processing units, graphics processing units and field-programmable gate arrays are implemented and their performances compared. Finally, MP-FAST-based image classification is evaluated using a multi-layer perceptron on various data sets, including both synthetic and experimental data. age Bragg Spot Finder (BSF): a new machine-learning-aided approach to deal with spot finding for rapidly filtering diffraction pattern images By journals.iucr.org Published On :: 2024-04-26 Macromolecular crystallography contributes significantly to understanding diseases and, more importantly, how to treat them by providing atomic resolution 3D structures of proteins. This is achieved by collecting X-ray diffraction images of protein crystals from important biological pathways. Spotfinders are used to detect the presence of crystals with usable data, and the spots from such crystals are the primary data used to solve the relevant structures. Having fast and accurate spot finding is essential, but recent advances in synchrotron beamlines used to generate X-ray diffraction images have brought us to the limits of what the best existing spotfinders can do. This bottleneck must be removed so spotfinder software can keep pace with the X-ray beamline hardware improvements and be able to see the weak or diffuse spots required to solve the most challenging problems encountered when working with diffraction images. In this paper, we first present Bragg Spot Detection (BSD), a large benchmark Bragg spot image dataset that contains 304 images with more than 66 000 spots. We then discuss the open source extensible U-Net-based spotfinder Bragg Spot Finder (BSF), with image pre-processing, a U-Net segmentation backbone, and post-processing that includes artifact removal and watershed segmentation. Finally, we perform experiments on the BSD benchmark and obtain results that are (in terms of accuracy) comparable to or better than those obtained with two popular spotfinder software packages (Dozor and DIALS), demonstrating that this is an appropriate framework to support future extensions and improvements. The PADF can be measured using X-ray or electron fluctuation diffraction data, which can be collected by scanning or flowing a structurally disordered sample through a focused beam. It is a natural generalization of established pair distribution methods, which do not provide angular information. The software package pypadf provides tools to calculate the PADF from fluctuation diffraction data. The package includes tools for calculating the intensity correlation function, which is a necessary step in the PADF calculation and also the basis for other fluctuation scattering analysis techniques. age Neural networks for rapid phase quantification of cultural heritage X-ray powder diffraction data By journals.iucr.org Published On :: 2024-05-31 Recent developments in synchrotron radiation facilities have increased the amount of data generated during acquisitions considerably, requiring fast and efficient data processing techniques. Here, the application of dense neural networks (DNNs) to data treatment of X-ray diffraction computed tomography (XRD-CT) experiments is presented. Processing involves mapping the phases in a tomographic slice by predicting the phase fraction in each individual pixel. DNNs were trained on sets of calculated XRD patterns generated using a Python algorithm developed in-house. An initial Rietveld refinement of the tomographic slice sum pattern provides additional information (peak widths and integrated intensities for each phase) to improve the generation of simulated patterns and make them closer to real data. A grid search was used to optimize the network architecture and demonstrated that a single fully connected dense layer was sufficient to accurately determine phase proportions. This DNN was used on the XRD-CT acquisition of a mock-up and a historical sample of highly heterogeneous multi-layered decoration of a late medieval statue, called `applied brocade'. The phase maps predicted by the DNN were in good agreement with other methods, such as non-negative matrix factorization and serial Rietveld refinements performed with TOPAS, and outperformed them in terms of speed and efficiency. The method was evaluated by regenerating experimental patterns from predictions and using the R-weighted profile as the agreement factor. age Determination of the average crystallite size and the crystallite size distribution: the envelope function approach EnvACS By journals.iucr.org Published On :: 2024-09-17 A procedure is presented to exactly obtain the apparent average crystallite size (ACS) of powder samples using standard in-house powder diffraction experiments without any restriction originating from the Scherrer equation. Additionally, the crystallite size distribution within the sample can be evaluated. To achieve this, powder diffractograms are background corrected and long-range radial distribution functions G(r) up to 300 nm are calculated from the diffraction data. The envelope function fenv of G(r) is approximated by a procedure determining the absolute maxima of G(r) in a certain interval (r range). Fitting of an ACS distribution envelope function to this approximation gives the ACS and its distribution. The method is tested on diffractograms of LaB6 standard reference materials measured with different wavelengths to demonstrate the validity of the approach and to clarify the influence of the wavelength used. The latter results in a general description of the maximum observable average crystallite size, which depends on the instrument and wavelength used. The crystallite site distribution is compared with particle size distributions based on transmission electron microscopy investigations, providing an approximation of the average number of crystallites per particle. Crystal images of single crystals of the β-form of l-glutamic acid are captured in situ during their growth at a relative supersaturation of 1.05 using transmission optical microscopy. The crystal growth rates estimated for both the {101} capping and {021} prismatic faces through image processing are consistent with those determined using reflection light mode [Jiang, Ma, Hazlehurst, Ilett, Jackson, Hogg & Roberts (2024). Cryst. Growth Des. 24, 3277–3288]. The growth rate in the {010} face is, for the first time, estimated from the shadow widths of the {021} prismatic faces and found to be typically about half that of the {021} prismatic faces. Analysis of the 3D shape during growth reveals that the initial needle-like crystal morphology develops during the growth process to become more tabular, associated with the Zingg factor evolving from 2.9 to 1.7 (>1). The change in relative solution supersaturation during the growth process is estimated from calculations of the crystal volume, offering an alternative approach to determine this dynamically from visual observations. Just last month, Microsoft Corp. (MSFT:NASDAQ) announced a deal with Constellation Energy Group (CEG:NYSE) to restart and buy all of the power from one of the shut-down reactors at its infamous Three Mile Island plant in Pennsylvania and the Biden administration also announced a plan to restart the Palisades plant in Michigan. "Biden has called for a tripling of U.S. nuclear power capacity to fuel energy demand that is accelerating in part due to expansion of power-hungry technologies like artificial intelligence and cloud computing," Valerie Volcovici wrote for Reuters on Oct. 8. The administration also wants to develop small nuclear reactors (SMRs) for certain applications. All of this is putting the metal needed to power nuclear energy, uranium, front and center. Prices for the element have started rising, with nuclear fuel trading at US$83.30 per pound last Thursday, a level not seen since 2007, according to a report by Daily Finland on Friday. Uranium prices are expected to move higher by the end of this quarter, when Trading Economics' global macro models and analyses forecast uranium to trade at US$84.15 per pound, Nuclear Newswire reported on Oct. 3. In another year, the site estimates that the metal will trade at US$91.80 per pound. The Catalyst: Surging Demand The engine driving the prices is a "fundamental global shortage" of uranium driven by surging demand, said Andre Leibenberg, chief executive officer of Yellow Cake, which is focused on providing exposure to uranium's spot price. The demand is stemming not only from a growing recognition of nuclear power's role in the future energy mix, but also from its critical importance in supporting the AI boom and the development of data centers, he wrote in a company update last week, according to Mining Weekly. According to the report, Liebenberg noted that the primary mine supply of 140 million pounds was significantly trailing behind global demand of more than 180 million pounds a year. In the European Union, a "lack of clarity" about Russian uranium imports is holding back investment in new enrichment plants, according to Reuters. Russia supplied more than 25% of European and American enriched uranium before the start of the Ukraine war in February 2022, the report said. Since then, "the U.S. implemented a ban on imports of enriched uranium from Russia in August, with some exemptions, but in Europe, different countries have taken different approaches," muddying the waters. Complicating matters is a hint in September that Russian President Vladimir Putin might embargo exports of the vital element to the west. Citi, in a note to clients, said utilities have been stockpiling Russian uranium, but an embargo would make it "hard to replace" supplies of the metal in the next two years. "Russia supplies close to 12% of U3O8 (known as yellow cake), 25% of UF6 (uranium hexafluoride) and 35% of EUP (enriched uranium product) to international markets," the bank said, according to Forbes. "While the largest share of these supplies goes to China and in supplying nuclear reactors that were built by Russia's Rosatom, we believe that at-risk supplies are exports to the U.S. or Western Europe." The consequences of what could happen without more nuclear power can be seen in the U.K., where the number of reactors is shrinking. Four of five of them are expected to close in the next couple of years, which could "stretch the grid to the limit." "As Britain's reactor fleet shrivels, the amount of nuclear capacity will fall from six gigawatts (GW) today to just 1.2 GW by 2028 or soon after," Jonathan Leake and Matt Oliver wrote for The Telegraph last week. "Along with rising demand from power-hungry data centers and technologies of the future, it will make it even harder to keep the lights on when wind and solar generation is low." Small Nuclear Reactors (SMRs) SMRs are another possible solution for some medium-sized energy needs. They have been operational for dozens of years in submarines and other long-distance ocean-going craft. "They can be manufactured in factories and then rapidly erected on-site," Dominic Frisby wrote for his newsletter, The Flying Frisby, on Oct. 13. They are scalable, and that flexibility "aids manufacture, transportation, and installation while reducing construction time and costs." A 440-megawatt (MW) SMR would produce about 3.5 terawatt hours (TWh) of electricity per year, enough for 1.2 million homes, Frisby noted. SMRs produce electricity that can easily be adjusted to meet the constant, everyday needs of the grid (baseload), and they can also ramp up or down to follow changes in demand throughout the day, the author wrote. They spin in sync with the grid, so they help keep everything stable. "When they're running, they act like a steady hand, providing momentum that makes it easier to manage sudden changes in electricity supply or demand," he wrote. 'Bucket Loads of Power' Needed All of this equates for a bright future for the metal, he said. "Guess what? AI requires bucket loads of power," Frisby wrote. "That's why Microsoft recently agreed to pay Constellation Energy, the new owner of America's infamous nuclear power station, Three Mile Island, a sizeable premium for its energy. There is cheaper wind and solar power to be had in Pennsylvania, but it isn't as reliable as nuclear 24 hours a day. It's not just AI. The widespread political desire to rid ourselves of fossil fuels means the world needs electricity, and fast." Chris Temple, publisher of The National Investor, recently noted that with the Three Mile Island deal, "uranium/nuclear power is BACK!" "I've watched as the news has continued to point to uranium being in the early innings of this new bull market," Temple wrote. "Yet the markets have been yawning . . . until now." What follows are several uranium explorers and producers that could benefit from this upswing for investors looking to take advantage. Baselode Energy Corp. Baselode Energy Corp. (FIND:TSX.V; BSENF:OTCQB) controls 100% of about 273,000 hectares for exploration in the Athabasca Basin area in northern Saskatchewan, Canada.[OWNERSHIP_CHART-10321] The company said it discovered the ACKIO near-surface, high-grade uranium deposit in September 2021. ACKIO measures greater than 375 meters along strike, greater than 150 meters wide, and is comprised of at least 11 separate zones. Mineralization starts as shallow as 28 meters beneath the surface and continues down to about 300 meters depth beneath the surface, with the bulk of mineralization occurring in the upper 120 meters, Baselode said. ACKIO remains open to the west and south and along the Athabasca sandstone unconformity to the east and south. Earlier this month, the company reported positive uranium assay results from three drill holes of its 2024 drill program at ACKIO. Notably, drill hole AK24-119 intersected 0.28% U3O8 over 21.0 meters, including a high-grade section of 1.55% U3O8 over 1.5 meters at a depth of 141 meters. While drill hole AK24-118 returned 0.59% U3O8 over 8.5 meters, including 1.25% U3O8 over 1.5 meters at a depth of 153 meters. "These results strengthen our confidence in ACKIO," Chief Executive Officer James Sykes said in a release. "It's remarkable that, just over three years after discovering ACKIO, we're still achieving better-than-expected grades and widths." Baselode expects further assay results from the remaining 40 drill holes to be released after quality review and approval. David Talbot, Managing Director at Red Cloud Securities, noted in a September 17 report that drilling at ACKIO "continued to expand the mineralized footprint at Pods 1, 6, and 7," highlighting that "thirteen holes reported composite intervals of anomalous radioactivity between 11m and 42m in thickness." In his report, Talbot rated the stock as a Buy and further projected the potential for "8-10-12 million pounds of U3O8 at a grade of ~0.3% U3O8," which aligns with typical grades found in the southeastern part of the Athabasca Basin. According to Refinitiv, Baselode has institutions holding 23.26% with Alps Advisors holding the bulk of it with 17.94%, followed by Vident Investment Advisory LLC at 2.97%. Management and Insiders hold 1.59%. The rest is retail. The company has a market cap of CA$20.05 million, with 131.51 free float shares. It trades in the 52-week range between CA$0.10 and CA$0.61. Uranium Energy Corp. According to its website, Uranium Energy Corp. (UEC:NYSE AMERICAN) is America's "largest and fastest growing supplier of uranium."[OWNERSHIP_CHART-402] The company said it is advancing the next generation of low-cost, environmentally friendly in-situ recovery (ISR) mining uranium projects in the United States and high-grade conventional projects in Canada. It has two production-ready ISR hub and spoke platforms in South Texas and Wyoming. Additionally, Uranium Energy Corp. said it has diversified uranium holdings with one of the largest physical uranium portfolios of U.S. warehoused U3O8; a major equity stake in Uranium Royalty Corp., the only royalty company in the sector; and a Western Hemisphere pipeline of resource stage uranium projects. Most recently, the company announced it was expanding its U.S. uranium production capacity by acquiring Rio Tinto Plc.'s Sweetwater Plant and a portfolio of Wyoming uranium assets. On September 25, Temple of The National Investor noted that UEC was "upgraded back to Buy" following recent uranium market news. He pointed to UEC's acquisition of the Wyoming uranium assets as a catalyst, emphasizing that uranium is "in the early innings of this new bull market." Jeff Clark of The Gold Advisor, in his September 26 update, called the acquisition a "significant move," noting that it consolidated a large portfolio of uranium assets under UEC's control, positioning the company for rapid growth. He also highlighted the company's strategic advantage with "53,000 additional acres for exploration," reinforcing UEC's potential to ramp up production. According to Reuters, Uranium Energy has a market cap of US$3.48 billion and 411.41 million shares outstanding. It trades in a 52-week range of US$4.06 and US$8.66. About 2% of UE is help by management and insiders, Reuters noted. The largest portion, 77.58%, is held by institutional investors. The rest is in retail. Terra Clean Energy Corp. Formerly Tisdale Clean Energy Corp., Terra Clean Energy Corp. (TCEC:CSE; TCEFF:OTC; T1KC:FSE), a Canadian-based uranium exploration and development company, is currently developing the South Falcon East uranium project, which holds a 6.96-million-pound inferred uranium resource within the Fraser Lakes Zone B uranium/thorium deposit, located in the Athabasca Basin region of Saskatchewan.[OWNERSHIP_CHART-10935] Representing a portion of Skyharbour Resources Ltd.'s existing South Falcon Project, Terra Clean Energy's project covers approximately 12,464 hectares and lies 18 kilometers outside the Athabasca Basin, approximately 50 kilometers east of the Key Lake Mine. Recently, the company announced a comprehensive exploration program set for Winter 2025 at its South Falcon East Uranium Project. The work will focus on extending the mineralized footprint at the Fraser Lakes B Uranium Deposit and includes about 2,000 meters of infill and step-out drilling designed to verify existing mineralized zones and identify additional targets. In a release, Chief Executive Officer Alex Klenman described the initiative as "a unique setup for a Canadian microcap, offering multiple paths to significant value creation." This US$1.5 million project will involve TerraLogic Exploration Inc., operating out of SkyHarbour's McGowan Lake Camp with helicopter support. According to Reuters, management and insiders hold 4.62% of Terra Clean Energy. Of those, Alex Klenman holds the most, with 4.37%. Strategic Investors hold 12.03%, with Planet Ventures Inc holding the most at 7.40%. The rest is retail. Terra Clean Energy has a market cap of CA$2.98 million and a 52-week range of CA$0.05 to CA$0.22. North Shore Uranium Ltd. North Shore Uranium Ltd. (NSU:TSX) said it is working to become a major force in exploration for economic uranium deposits at the eastern margin of the Athabasca Basin.[OWNERSHIP_CHART-10945] The company said it is running exploration programs at its Falcon and West Bear properties and evaluating opportunities to complement its portfolio of uranium properties. Falcon consists of 15 mineral claims, the company said. Four of them comprise 12,791 hectares and are 100%-owned by the company. The remaining 11 claims totaling 2,908 hectares are subject to an option agreement with Skyharbour Resources Ltd. Under the terms of the option agreement, North Shore has the option to earn up to 100% interest in the 11 claims by completing certain payments. Earlier this month, the company announced details of its target generation efforts at its Falcon uranium project at the eastern margin of Saskatchewan's Athabasca Basin. The company said it has identified 36 uranium targets across three zones. "We have a great pipeline of targets to choose from for our next drill program at Falcon," said President and Chief Executive Officer Brooke Clements. "Our Zone 2 has attracted the interest of uranium explorers in the past, and we believe there is potential to make a significant uranium discovery using new data and interpretation." Earlier this month, North Shore announced it had received a Crown Land Work permit for the full 55,700-hectare Falcon project. Issued by the Saskatchewan Ministry of Environment, it authorizes the company to conduct mineral exploration activities, including prospecting and ground geophysics, trail and drill site clearing, line cutting, the drilling of up to 75 exploration drill holes, and the storage of drill core. The permit expires in July 2027. Insiders and founding investors own approximately 45% of the issued and outstanding shares. Clements himself owns 3.6% or 1.33M shares, Director Doris Meyer has 2.11% or 0.78M shares, and Director James Arthur holds 1.58% or 0.58M shares. According to North Shore, 14.92M shares (40.5%) held by six founding investors are subject to a voluntary pooling agreement that restricts the disposition of these shares before October 19, 2026. Most of the rest is with retail, as the institutional holdings are minor. North Shore has 36.84M outstanding shares and currently has a market cap of CA$1.47 million. (SYH:TSX.V; SYHBF:OTCQX; SC1P:FSE) has an extensive portfolio of uranium exploration projects in Canada's Athabasca Basin, with 29 projects, 10 of which are drill-ready, covering over 1.4 million acres of mineral claims. In addition to being a high-grade uranium exploration company, Skyharbour utilizes a prospect generator strategy by bringing in partner companies to advance its secondary assets.[OWNERSHIP_CHART-6026] In an updated research note on July 24, Analyst Sid Rajeev of Fundamental Research Corp. wrote that Skyharbour "owns one of the largest portfolios among uranium juniors in the Athabasca Basin." "Given the highly vulnerable uranium supply chain, we anticipate continued consolidation within the sector," wrote Rajeev, who reiterated the firm's Buy rating and adjusted its fair value estimate from CA$1.16 to CA$1.21 per share. "Additionally, the rapidly growing demand for energy from the AI industry is likely to accelerate the adoption of nuclear power, which should, in turn, spotlight uranium juniors in the coming months." Skyharbour acquired from Denison Mines, a large strategic shareholder of the company, a 100% interest in the Moore Uranium Project, which is located 15 kilometers east of Denison's Wheeler River project and 39 kilometers south of Cameco's McArthur River uranium mine. Moore is an advanced-stage uranium exploration property with high-grade uranium mineralization at the Maverick Zone, including highlight drill results of 6.0% U3O8 over 5.9 meters, including 20.8% U3O8 over 1.5 meters at a vertical depth of 265 meters. Adjacent to the Moore Uranium Project is Skyharbour's Russell Lake Uranium Project optioned from Rio Tinto, which hosts historical high-grade drill intercepts over a large property area with robust exploration upside potential. The 73,294-ha Russell Lake Uranium Property is strategically located in the central core of the Eastern Athabasca Basin of northern Saskatchewan. Skyharbour has recently discovered high-grade uranium mineralization in a new zone at Russell and is carrying out an additional 7-8,000-meter drill campaign across both Russell and Moore. Management, insiders, and close business associates own approximately 5% of Skyharbour. According to Reuters, President and CEO Trimble owns 1.6%, and Director David Cates owns 0.70%. Institutional, corporate, and strategic investors own approximately 55% of the company. Denison Mines owns 6.3%, Rio Tinto owns 2.0%, Extract Advisors LLC owns 9%, Alps Advisors Inc. owns 9.91%, Mirae Asset Global Investments (U.S.A) L.L.C. owns 6.29%, Sprott Asset Management L.P. owns 1.5%, and Incrementum AG owns 1.18%, Reuters reported. There are 182.53 million shares outstanding with 178 million free float traded shares, while the company has a market cap of CA$89.44 million and trades in a 52-week range of CA$0.31 and CA$0.64. (SASK:TSX.V; SASKF:OTCMKTS) is a Canadian mineral company engaged in the acquisition, exploration, and development of uranium assets with a portfolio including three 100%-owned post-discovery uranium projects (the Angilak Project located in Nunavut, and CMB Discoveries in Labrador hosting historical resource estimates of 43.3 million pounds and 14.5 million pounds U3O8 respectively, and the newly discovered basement-hosted GMZ high-grade uranium discovery located in the Athabasca Basin).[OWNERSHIP_CHART-11007] In addition, the company said it holds the largest cumulative prospective exploration land package (more than 8.5 million acres) in two of the world's most prominent basins for uranium discoveries. Technical Analyst Maund considers Atha Energy to be "THE top play in the uranium sector" and has an Immediate Strong Buy rating on it, he wrote in the previously mentioned Oct. 17 report. The company's 3-, 13- and 26-month charts indicate its stock price had been in a bear market since trading began until September, when it had an upwave or preliminary breakout. This, along with other indicators, including positive accumulation-distribution convergence and high volume, suggest another upleg is expected soon, he said. "Given the outlook for the uranium price and what Atha Energy has going for it, its stock is astoundingly cheap after its persistent downtrend this year," Maund wrote. According to Refinitiv, 10 management and insiders own 16.44% of Atha Energy. The Top 5 are Timothy Young with 6.32%, Matthew Mason with 5.8%, Atha Chairman Michael Castanho with 1.16%, and Atha Director Sean Kallir with 0.9%. Seven institutional investors together hold 9.38%. This comes as the need for more net-zero power is sparking a rebirth of the nuclear industry.Skyharbour Resources Ltd. (SYH:TSX.V; SYHBF:OTCQX; SC1P:FSE) announced that it has completed its earn-in requirements for a 51% interest at its co-flagship Russell Lake Uranium Project in the central core of Canada's Eastern Athabasca Basin in Saskatchewan. The company and Rio Tinto have formed a joint venture (JV) to further explore the property, with Skyharbour holding 51% ownership interest and Rio Tinto holding 49%. This summer, Skyharbour announced that in the first phase of drilling it had found what was historically the best uranium intercept mineralization at the project when hole RSL24-02 at the recently identified Fork Target returned a 2.5-meter-wide intercept of 0.721% U3O8 at a relatively shallow depth of 338.1 meters, including 2.99% U3O8 over 0.5 meters at 339.6 meters. The second phase of drilling included three holes totaling 1,649 meters, with emphasis "at the MZE (M-Zone Extension) target, approximately 10 km northeast of the Fork target, identified prospective faulted graphitic gneiss accompanied by anomalous sandstone and basement geochemistry," Skyharbour said. "The discovery of multi-percent, high-grade, sandstone-hosted uranium mineralization at a new target is a major breakthrough in the discovery process at Russell — something that hasn't been seen before at the project with the potential to quickly grow with more drilling," President and Chief Executive Officer Jordan Trimble said at the time. ANT Survey, Upcoming Drilling Program The company also announced on Thursday that it had completed an Ambient Noise Tomography (ANT) survey in preparation for further drilling at the Russell Lake Project, set to commence in the fall. The survey used Fleet Space Technologies' Exosphere technology to acquire 3D passive seismic velocity data over the highly prospective Grayling and Fork target areas, where previous drilling has intersected high-grade uranium mineralization. "The ANT technology has been successfully employed in mapping significant sandstone and basement structures and associated alteration zones related to hydrothermal fluids pathways in the Athabasca Basin," the company said. Results from the survey will be used to further refine drill targets for the upcoming drilling program. Skyharbour is fully funded and permitted for the follow-up fall drill campaign consisting of approximately 7,000 metres of drilling at its main Russell and Moore Projects, with 2,500 meters of drilling at Moore and 4,500 meters of drilling at Russell. A Great Neighborhood Russell Lake is a large, advanced-stage uranium exploration property totaling 73,294 hectares strategically located between Cameco's Key Lake and McArthur River projects and Denison's Wheeler River Project to the west, and Skyharbour's Moore project to the east. "Skyharbour's acquisition of a majority interest in Russell Lake creates a large, nearly contiguous block of highly prospective uranium claims totaling 108,999 hectares between the Russell Lake and the Moore uranium projects," the company said. Most of the historical exploration at Russell Lake was conducted before 2010, prior to the discovery of several major deposits in/around the Athabasca Basin, Skyharbour said. Notable exploration targets on the property include the Grayling Zone, the M-Zone Extension target, the Little Man Lake target, the Christie Lake target, the Fox Lake Trail target and the newly identified Fork Zone target. "More than 35 kilometers of largely untested prospective conductors in areas of low magnetic intensity also exist on the property," the company noted. In an updated research note in July, Analyst Sid Rajeev of Fundamental Research Corp. wrote that Skyharbour "owns one of the largest portfolios among uranium juniors in the Athabasca Basin." "Given the highly vulnerable uranium supply chain, we anticipate continued consolidation within the sector," wrote Rajeev, who rated the stock a Buy with a fair value estimate of CA$1.21 per share. "Additionally, the rapidly growing demand for energy from the AI (artificial intelligence) industry is likely to accelerate the adoption of nuclear power, which should, in turn, spotlight uranium juniors in the coming months." The Catalyst: Uranium is 'BACK!' The growth of AI, new data centers, electric vehicle (EV) adoption, and the need for more net-zero power means more nuclear energy and the uranium needed to fuel it. Uranium prices are expected to move higher by the end of this quarter, when Trading Economics' global macro models and analyses forecast uranium to trade at US$84.15 per pound, Nuclear Newswire reported on Oct. 3. In another year, the site estimates that the metal will trade at US$91.80 per pound. Just last month, Microsoft Corp. (MSFT:NASDAQ) announced a deal with Constellation Energy Group (CEG:NYSE) to restart and buy all of the power from one of the shut-down reactors at its infamous Three Mile Island plant in Pennsylvania and the Biden administration also announced a plan to restart the Palisades plant in Michigan. Chris Temple, publisher of The National Investor, recently noted that with the Three Mile Island deal, "uranium/nuclear power is BACK!"[OWNERSHIP_CHART-6026] "I've watched as the news has continued to point to uranium being in the early innings of this new bull market," Temple wrote. "Yet the markets have been yawning . . . until now." Ownership and Share Structure Management, insiders, and close business associates own approximately 5% of Skyharbour. He was 91.; Credit: Charles Dharapak/AP The Associated Press | NPRSEASIDE, Calif. — Mike Gravel, a former U.S. senator from Alaska who read the Pentagon Papers into the Congressional Record and confronted Barack Obama about nuclear weapons during a later presidential run, has died. He was 91. Gravel, who represented Alaska as a Democrat in the Senate from 1969 to 1981, died Saturday, according to his daughter, Lynne Mosier. Gravel had been living in Seaside, California, and was in failing health, said Theodore W. Johnson, a former aide. Gravel's two terms came during tumultuous years for Alaska when construction of the trans-Alaska oil pipeline was authorized and when Congress was deciding how to settle Alaska Native land claims and whether to classify enormous amounts of federal land as parks, preserves and monuments. He had the unenviable position of being an Alaska Democrat when some residents were burning President Jimmy Carter in effigy for his measures to place large sections of public lands in the state under protection from development. Gravel feuded with Alaska's other senator, Republican Ted Stevens, on the land matter, preferring to fight Carter's actions and rejecting Stevens' advocacy for a compromise. In the end, Congress passed the Alaska National Interest Lands Conservation Act of 1980, a compromise that set aside millions of acres for national parks, wildlife refuges and other protected areas. It was one of the last bills Carter signed before leaving office. Gravel's Senate tenure also was notable for his anti-war activity. In 1971, he led a one-man filibuster to protest the Vietnam-era draft and he read into the Congressional Record 4,100 pages of the 7,000-page leaked document known as the Pentagon Papers, the Defense Department's history of the country's early involvement in Vietnam. Gravel reentered national politics decades after his time in the Senate to twice run for president. Gravel, then 75, and his wife, Whitney, took public transportation in 2006 to announce he was running for president as a Democrat in the 2008 election ultimately won by Obama. He launched his quest for the 2008 Democratic presidential nomination as a critic of the Iraq war. "I believe America is doing harm every day our troops remain in Iraq — harm to ourselves and to the prospects for peace in the world," Gravel said in 2006. He hitched his campaign to an effort that would give all policy decisions to the people through a direct vote, including health care reform and declarations of war. Gravel garnered attention for his fiery comments at Democratic forums. In one 2007 debate, the issue of the possibility of using nuclear weapons against Iran came up, and Gravel confronted then-Sen. Obama. "Tell me, Barack, who do you want to nuke?" Gravel said. Obama replied: "I'm not planning to nuke anybody right now, Mike." Gravel then ran as a Libertarian candidate after he was excluded from later Democratic debates. In an email to supporters, he said the Democratic Party "no longer represents my vision for our great country." "It is a party that continues to sustain war, the military-industrial complex and imperialism — all of which I find anathema to my views," he said. He failed to get the Libertarian nomination. Gravel briefly ran for the Democratic nomination for president in 2020. He again criticized American wars and vowed to slash military spending. His last campaign was notable in that both his campaign manager and chief of staff were just 18 at the time of his short-lived candidacy. "There was never any ... plan that he would do anything more than participate in the debates. He didn't plan to campaign, but he wanted to get his ideas before a larger audience," Johnson said. Gravel failed to qualify for the debates. He endorsed Vermont Sen. Bernie Sanders in the contest eventually won by now-President Joe Biden. Gravel was born Maurice Robert Gravel in Springfield, Massachusetts, on May 13, 1930. In Alaska, he served as a state representative, including a stint as House speaker, in the mid-1960s. He won his first Senate term after defeating incumbent Sen. Ernest Gruening, a former territorial governor, in the 1968 Democratic primary. He was beloved for riveting and yet unsentimental depictions of the American West in books like Lonesome Dove, as well as for tales of family drama including Terms of Endearment. In a statement, his representative Amanda Lundberg said McMurtry "passed away last night, on March 25 of heart failure at 84 years old surrounded by his loved ones who he lived with including long time writing partner Diana Ossana, his wife Norma Faye and their 3 dogs." In all, McMurtry wrote more than 30 novels as well as over a dozen non-fiction works that spanned memoir, history and essays. He also wrote over 20 screenplays and television scripts. McMurtry was also famous for his bookstore, Booked Up in Archer City, Texas. Even after selling off more than half of his holdings in 2012, he still had about 200,000 books between his private collection and the store. When he won an Oscar in 2006 for the screenplay adaption of E. Annie Proulx' short story Brokeback Mountain, which he co-wrote with longtime writing partner Diana Ossana, he thanked booksellers. "From the humblest paperback exchange to the masters of the great bookshops of the world," he said, "all are contributors to the survival of the culture of the book, a wonderful culture, which we mustn't lose." Filmmakers were drawn to McMurtry's work; his books Hud, The Last Picture Show and Terms of Endearment were all made into films. Lonesome Dove, which earned him the Pulitzer in 1985, became a successful TV miniseries in 1989, starring Robert Duvall and Tommy Lee Jones. Born in 1936 on a Texas ranch, McMurtry came to his love of the West through his family. His grandfather broke horses, and his father raised cattle. "The West is mostly a very beautiful place," he told All Things Considered in 2014. "There are all those lovely spaces. There are all those running horses. It's a poetic imagery and it's been there for a long time." But he wanted to scour that landscape of sentimental nostalgia for cowboys, he added. "To me it was hollow and I think it was hollow for my father, although he might not have ever brought that to his conscious mind. He totally loved cowboys and so did most of the cowboys we worked with and that got him through his life. Prior to his current role as a professor of oceanography at Oregon State University, Spinrad served as NOAA's top scientist under President Obama and the U.S. representative to the United Nations' Intergovernmental Oceanographic Commission. The nomination comes at a difficult moment in NOAA's history. The agency has been without an official, Senate-confirmed leader since former President Donald Trump took office in January 2017, after his two nominees to lead the agency failed to garner enough support to win a full vote before the Senate. If Spinrad manages to win over the Senate, he will have to contend with a challenge beyond the agency's already-rigorous scientific mandate: restoring public confidence in a traditionally apolitical agency marred by political scandal. In September 2019, then-President Trump wrongly said Alabama was in the projected path of Hurricane Dorian. He continued to reassert the claim for several days, including during an Oval Office briefing on the storm — in which he displayed what appeared to be an official National Weather Service map in which the storm's projected path was extended to Alabama by someone using a black marker. After a National Weather Service office in Birmingham put out a tweet correctly stating that Alabama would not feel the effects of the storm, NOAA published an unsigned defense of the president's claims and rebuking its professional staff who posted the message. Dan Sobien, then-president of the National Weather Service Employees Organization, said at the time that "the hard working employees of the NWS had nothing to do with the utterly disgusting and disingenuous tweet sent out by NOAA management." If confirmed, Spinrad will lead a 12,000-person agency charged with a diverse portfolio that spans daily weather forecasts, climate monitoring, fisheries management and coastal restoration. In a statement, the Environmental Defense Fund's Eric Schwaab applauded Spinrad's nomination, saying that NOAA's workers "couldn't ask for a better leader to restore scientific integrity and honor the agency's mission." Biden, whose administration has made climate action a central focus, has proposed the largest budget in NOAA's history — $6.9 billion, a $1.5 billion increase over the 2021 budget allocated by Congress. age Obesity Drug's Promise Now Hinges On Insurance Coverage By www.scpr.org Published On :: Tue, 06 Jul 2021 04:00:11 -0700 Yuki Noguchi | NPRWhen a promising new drug to treat obesity was approved by the Food and Drug Administration for sale in the U.S. last month, it was the first such treatment to gain approval since 2014. In clinical trials, weekly injections of semaglutide — or Wegovy, as it's been branded -- helped people drop an average of 15% of their body weight. That's an average of about 34 pounds over 16 months, before their weight plateaued, a far greater weight loss, obesity specialists say, than achieved with other drugs on the market. At least as important, Wegovy raised none of the alarm bells with the FDA or obesity doctors that it might trigger serious side effects of the sort experienced by some people taking fen-phen or some previous medical treatments for obesity. But with a price tag for Wegovy of $1,000 to $1,500 a month, a very big question remains: Will insurers cover its significant cost for the many millions of people like Marleen Greenleaf, who might benefit? Greenleaf grew up on the island of Trinidad, where her entire family paid little heed to what they ate and paid a high medical price, she says: "My husband has diabetes, my sister has diabetes, my brother has diabetes." Since then, she's tried — and failed — at numerous diets, says Greenleaf, now 58 and an administrator at a charter school in Washington, D.C. Then, in 2018, she signed up for the clinical trial of a new drug — a once-weekly shot that changes the way her brain signals hunger. A drug that finally stops her cravings She noticed the change soon after her first injection of Wegovy: "For me, there was something that triggered in my brain to tell me that I was not hungry," she says. No more fierce cravings for the chocolate chip cookies she adores. Without the cravings she was able to slow down and reconsider the foods she'd been reaching for. "I also wanted to eat healthier," she says. "I was looking at options, reading labels, looking at the calories — not just the calories, but also the sugar." Over the 68-week research trial, Greenleaf dropped 40 pounds. Her blood pressure fell, which meant she qualified to donate her kidney to her husband, who was on dialysis. "It was one of the best gifts of life that I could have ever given," she says. But after that study ended, Greenleaf regained some of the weight. Wegovy is considered a long-term, possibly lifelong medication to treat chronic obesity. In the pre-marketing clinical studies, weight loss topped out at a total average weight loss of 15-18%, even as people continued to take the drug. And, as was the case with Greenleaf, once they stopped getting the weekly injections some of that weight came back. Now, Greenleaf wants to resume the Wegovy shots. "My only challenge actually is getting the insurance company to approve it," she says. Reimbursement for obesity drugs' cost is patchy Insurance coverage, it turns out, is a giant question — not just with Wegovy, but with obesity drugs in general. Some private insurers do include some prescription obesity drugs in the list of medicines they'll cover; it's too early to tell whether Wegovy will make those lists. Many doctors and patients are optimistic, because it is a higher dose of an existing diabetes medication called Ozempic, which is often covered by insurers. A few select state Medicaid programs will cover medications that treat obesity, in some circumstances. But, significantly, Medicare does not cover obesity drugs — and many private insurers typically follow Medicare's lead. Yet the demand for a good treatment is there, says Dr. Fatima Cody Stanford, a leading obesity researcher at Harvard. She was not involved in conducting the Wegovy clinical trial, but closely followed it. "I'm excited about it," she says, because of the dramatic weight loss. The drug acts on the brain so people eat less and store less of what they eat. That helps address the excess weight as well as helping with numerous related diseases of the liver or heart, for example. Why the FDA has been slow to approve obesity treatments There is a long history of drugs that have looked like promising treatments for obesity, then failed. Decades ago, amphetamines, were prescribed, until their addictive properties became apparent. In the 1990s, the combination of fenfluramine and phentermine — administered as the diet drug fen-phen — was heavily marketed, only to later be pulled from the market for causing heart valve problems. Those experiences and others have made physicians skeptical. "In obesity medicine field, we've learned to be cautiously optimistic each time we have a new medication that looks promising," says Dr. Ihuoma Eneli, director of the Center for Healthy Weight and Nutrition at Nationwide Children's Hospital in Columbus, Ohio, who was not involved in the study of Wegovy. So far, Eneli does not see any obvious concerns with the class of drugs that includes Wegovy, and calls the results so far "very promising." Wegovy is similar to another drug made by Novo Nordisk — Saxenda — which has been on the market since 2014, and which Eneli occasionally prescribes to her pediatric patients who are struggling with obesity. In clinical research studies, the primary side effects reported after taking Wegovy affected the digestive system: nausea, diarrhea, vomiting, constipation, abdominal pain or intestinal infections. Eneli says such side effects and their frequency are milder than the problems that have arisen in the past. That good safety profile may mean the drug is "less likely to come up with unanticipated risks," she says. But, the new drug will be of little use, she and other doctors who treat obesity say, if it's not also affordable for patients. "Before I even bring up that drug with my patients, I'm looking to see which insurance they are having on the left side of my screen — because that will determine whether I bring it up," Stanford, the Harvard physician, says. "If it's out of reach, like I said, I won't bring it up." Stanford says her patients on existing obesity medications do extraordinary things to keep their coverage so they can afford to stay on the drugs. "Several nurses here at the hospital that are my patients stayed working — they were supposed to retire — so they could stay on their injectable medication," Stanford says,"because that's how beneficial it was to them." Why some are willing to pay out of pocket Some people, like David Scheesley, 42, says he would consider paying for Wegovy, even if he had to pay the full sticker price. The Hanford, Calif., correctional officer has tried since 2019 to lose weight on various diets — low-fat, all-meat, all-vegetable — without success. His weight has led to other health concerns — with his blood fats and his heart — which makes Scheesley think of his 5-year-old son. "I want to see him for a lot of years; I don't want to have a stroke," he says. "I don't want to have diabetes. I want to be there for him. So, for me personally, that [monetary cost] is not astronomical, if it can give me some more time." Novo Nordisk, the company that makes Wegovy, is in talks with insurers, and acknowledges that ensuring health insurance coverage of its drug is critical. The challenge, says Douglas Langa, executive vice president of Novo Nordisk North America, is getting doctors, patients, and politicians to recognize obesity as a disease — and that therefore insurance should cover the cost of medicine to treat it. "There's a medical component to [obesity] that needs to be recognized; this is a disease state like we should be treating any other disease state," Langa says. He says about 40% of private insurers cover Saxenda, the similar weight-loss medication the company makes. Langa tells insurance companies this, making the case for why prescriptions for Wegovy should be covered. His company is also heavily lobbying Congress to pass legislation to allow Medicare to cover obesity medications. It makes sense from a financial perspective, he argues, because obesity is the root disease underlying so many other diseases. "We do believe insurers understand that [untreated obesity] is a gateway into 60 other health conditions," Langa says. The need is hard to ignore, he adds.
