Synthesis and crystal structure of (1,10-phenanthroline-κ2N,N')[2-(1H-pyrazol-1-yl)phenyl-κ2N2,C1]iridium(III) hexafluoridophosphate with an unknown number of solvent molecules

The cationic complex in the title compound, [Ir(C9H7N2)2(C12H8N2)]PF6, comprises two phenylpyrazole (ppz) cyclometallating ligands and one 1,10-phenanthroline (phen) ancillary ligand. The asymmetric unit consists of one [Ir(ppz)2(phen)]+ cation and one [PF6]− counter-ion. The central IrIII ion is six-coordinated by two N atoms and two C atoms from the two ppz ligands as well as by two N atoms from the phen ligand within a distorted octahedral C2N4 coordination set. In the crystal structure, the [Ir(ppz)2(phen)]+ cations and PF6− counter-ions are connected with each other through weak intermolecular C—H...F hydrogen bonds. Additional C—H...π interactions between the rings of neighbouring cations consolidate the three-dimensional network. Electron density associated with additional disordered solvent molecules inside cavities of the structure was removed with the SQUEEZE procedure in PLATON [Spek (2015). Acta Cryst. C71, 9–18]. The given chemical formula and other crystal data do not take into account the unknown solvent molecule(s). The title compound has a different space-group symmetry (C2/c) from its solvatomorph (P21/c) comprising 1.5CH2Cl2 solvent molecules per ion pair.

In the first reported crystal structure involving the potential ligand N,N',N''-tris(pyridin-2-yl)benzene-1,3,5-tricarboxamide, C24H18N6O3, intermolecular N—H...O hydrogen bonds link the molecules via their amide groups into slanted ladder-like chains, in which the uprights of the ladder are formed by the hydrogen-bonding interactions and the benzene ring cores of the molecules act as the rungs of the ladder. Only two of the three amide groups in the molecule are involved in hydrogen bonding and this influences the degree of out-of-plane twisting at each amide group, with the twist being more significant for those amide groups participating in hydrogen bonds.

In small-molecule single-crystal structure determination, we now have at our disposal an inspiring range of fantastic diffractometers with better, brighter sources, and faster, more sensitive detectors. Polymorphism and phase transformation in the dimethyl sulfoxide solvate of 2,3,5,6-tetra­fluoro-1,4-di­iodo­benzene

A new polymorph (form II) is reported for the 1:1 dimethyl sulfoxide solvate of 2,3,5,6-tetra­fluoro-1,4-di­iodo­benzene (TFDIB·DMSO or C6F4I2·C2H6SO). The structure is similar to that of a previously reported polymorph (form I) [Britton (2003). Acta Cryst. E59, o1332–o1333], containing layers of TFDIB mol­ecules with DMSO mol­ecules between, accepting I⋯O halogen bonds from two TFDIB mol­ecules. Re-examination of form I over the temperature range 300–120 K shows that it undergoes a phase transformation around 220 K, where the DMSO mol­ecules undergo re-orientation and become ordered. The unit cell expands by ca 0.5 Å along the c axis and contracts by ca 1.0 Å along the a axis, and the space-group symmetry is reduced from Pnma to P212121. Refinement of form I against data collected at 220 K captures the (average) structure of the crystal prior to the phase transformation, with the DMSO mol­ecules showing four distinct disorder com­ponents, corresponding to an overlay of the 297 and 120 K structures. Assessment of the inter­molecular inter­action energies using the PIXEL method indicates that the various orientations of the DMSO mol­ecules have very similar total inter­action energies with the molecules of the TFDIB framework. The phase transformation is driven by inter­actions between DMSO mol­ecules, whereby re-orientation at lower temperature yields significantly closer and more stabilizing inter­actions between neighbouring DMSO mol­ecules, which lock in an ordered arrangement along the shortened a axis.

'Lost in Space' robot designer Robert Kinoshita dies at 100

Robert Kinoshita, the Los Angeles native who designed the iconic robots from "Lost in Space" and "Forbidden Planet," has passed away. He was 100 years old.

Konishita died Dec. 9 at a Torrance nursing home, according to the Hollywood Reporter, citing family friend Mike Clark. His creations included "Forbidden Planet's" Robby the Robot, the B9 robot from "Lost in Space," Tobor from "Tobor the Great" and more. Kinoshita also created "Lost in Space's" iconic flying-saucer-shaped Jupiter 2 spaceship.

Kinoshita built the original miniature prototype of Robby the Robot out of wood and plastic by combining several different concepts, according to the Reporter; the Rafu Shimpo reported that he struggled with the design.

"I thought, what the hell. We’re wasting so much time designing and drawing one sketch after another. I said to myself, I’m going to make a model," Kinoshita told the Rafu Shimpo in a 2004 interview. "Then one day, the art director sees the model. He says, ‘Give me that thing.’ He grabbed it and ran. ... Ten minutes later, he comes running back and puts the model back on my desk and says, ‘Draw it!’"

The 1956 classic sci-fi movie "Forbidden Planet" — based on Shakespeare's "The Tempest" — went on to be nominated for a special effects Oscar.

Kinoshita later served as art director on the 1960s sci-fi TV series "Lost in Space," creating the arm-flailing robot — named B9 — who delivered the classic line "Danger! Danger, Will Robinson!" That robot received as much fan mail as the actual humans on the show, according to the Reporter.

The "Lost in Space" robot even inspired a B9 Robot Builders Club, featured in Forbes. Kinoshita sent a message in 2000 to the club, thanking them for their support for the robot he originally nicknamed "Blinky."

"I'm truly flabbergasted and honored by your support for 'Blinky!' It's a well-designed little beauty," Kinoshita wrote. "Your thoughtful remembrance is something we designers seldom are lucky enough to receive."

Kinoshita described the thought process behind its design in a 1998 interview.

"You're laying in bed, and something comes to you," he said. "Until, finally, you get to a point where you say, 'This could work,' 'OK, let's see what the boss man says.' And you present it to him."

He told the Rafu Shimpo that he tried to create his robots to disguise the fact that there was a person inside. "I tried to camouflage it enough so you’d wonder where the hell the human was," he said.

Both the Japanese-American Kinoshita and his wife, Lillian, were sent to an Arizona internment camp during World War II, though they were able to get out before the end of the war and moved to Wisconsin, according to the Reporter.

While in Wisconsin, Kinoshita learned industrial design and plastic fabrication, designing washing machines for the Army and Air Force before returning to California, according to the Rafu Shimpo.

Kinoshita said that he had to overcome racial prejudice to break into working in Hollywood.

Kinoshita attributed his long life to clean living — along with daily doses of apple cider vinegar, family friend Clark told the Reporter.

Kinoshita also worked as a designer and art director on numerous classic TV shows, including "Kojak," "Barnaby Jones," "Hawaii Five-O," "Bat Masterson," "Sea Hunt," "Tombstone Territory," "Star Trek" creator Gene Roddenberry's "Planet Earth" and more, according to his IMDB. His last TV show was 1984's "Cover Up."

Kinoshita grew up in Boyle Heights, according to the Reporter, attending Maryknoll Japanese Catholic School, Roosevelt High School and USC's School of Architecture. His career began with work on 1937's "100 Men and a Girl." Kinoshita graduated cum laude from USC, according to the Rafu Shimpo.

Director Joseph Kahn tells Deadline, "They put these disclaimers on so kids so don’t confuse our super-violent film with their Power Rangers brand. There are no hard feelings. We signed contracts. Previously: A Power Rangers short film went viral this week, starring Katee Sackhoff (Battlestar Galactica) and James Van Der Beek (Dawson!) in a Tarantino-meets-sci-fi take on a dark future for the formerly Mighty Morphin'.

The company behind the 22-year history of the kids/toy icons issued takedown requests, leading to it being pulled from Vimeo and, on Thursday afternoon, a toned-down version being taken off YouTube. At the time, the video had received over 10 million views. It's still available to watch in this Facebook embed from the producer's Facebook page:

The film was directed by Joseph Kahn, best known as the music video director behind clips like Taylor Swift's "Blank Space" as well as videos for Britney Spears, U2 and others, and produced by Adi Shankar, who was also behind "The Grey" and "Dredd." Kahn co-wrote the film with Van Der Beek and lesser-known writer Dutch Southern.

This isn't the first quasi-legal fan film produced by Shankar. He's also done the same with Marvel Comics characters the Punisher and Venom. The Punisher film even had one of the actors who played the character on screen, Thomas Jane.

Kahn tells HitFix that Shankar came to him, offering him the chance to reboot an existing property.

One reason for the current legal contentiousness: The Power Rangers are set to hit the big screen next year in a Lionsgate film. Kahn says that, if that film was offered to him, he wouldn't be interested.

"I wouldn't even want to make 'Power Rangers: The Movie' for real," Kahn told HitFix. "Like if I had to make a 'Power Rangers' movie, this is it. It's 14 minutes long, and it's violent, and this is what I have in me. If they offered me the $200 million version, the PG-13 version, I literally wouldn't do it."

There's been a strong response to the video, but the response has been mixed — among the original Power Rangers. The original Green Ranger, actor Jason David Frank, said on Facebook that he was approached to be in the movie but declined. (The film's director denies this claim, saying that Frank must have confused them with another project.) Frank also criticized the adult content.

"I'm just a PG-13 guy," Frank said. "The cuss words, the drugs, all this other stuff, it doesn't fly with me." He says that he thinks the Power Rangers are still connected with kids, and he didn't like the idea of doing a dark and gritty version in that context. (Watch his video below.)

But the original Pink Ranger, actress Amy Jo Johnson, gave the film her stamp of approval, which Sackhoff enthusiastically responded to.

Sackhoff told the Nerdcore Movement site that she loves fan films.

"Doing a fan video is kind of the biggest honor you can give a franchise. It’s something that inspired you so much that you want to create," Sackhoff said.

Kahn defended "POWER/RANGERS" on Twitter as being protected due to it being a satire and also cited fair use laws and free speech. He argued that there are lots of other fan films and pointed out that YouTube showed more leniency, leaving it up.

"Every image in POWER/RANGERS is original footage," Kahn wrote in a series of tweets to Vimeo. "Nothing was pre-existing. There is no copyrighted footage in the short."

He also said that it's being given away for free and that Kahn paid for it himself.

"It is just as if I drew a pic of Power Rangers on a napkin and I gave it to my friend," Kahn wrote. "Is it illegal to give pic I drew of a character on a napkin to someone for free? No. The world is watching your actions right now."

In a touch of irony, the video had been selected as a Vimeo staff pick before the service took it down. They also responded to complaints about it being taken down, saying that they had to due to the Digital Millennium Copyright Act, and that they would restore the video immediately if the copyright claim was withdrawn, telling fans to complain to Power Rangers copyright holder Saban.

Despite the short film's success, hardcore Power Rangers fans are less sure about it, Shamus Kelley tells KPCC. Kelley created his own Rangers fan series, as well as co-hosting a Power Rangers podcast with over 300 episodes so far.

Casual fans are "far more open to the idea of dark/gritty because they see it as the show growing up with them," Kelley says. To hardcore fans, "it was either an abomination or the greatest unintended parody ever."

Kahn tells HitFix that the parody, of both Power Rangers and fan films in general, is definitely intended.

"The dark and gritty reboot thing is such a cliché that the intention was not only to make it dark and gritty but make it even darker and grittier than you could possibly imagine, hence the brains, the blood and the violence and the sex," Kahn said.

Kahn is set to answer questions about the film in a Reddit AMA on Friday at 2 p.m. We'll have to wait and see if there will be anywhere to watch it at the time.

Los Angeles Unified school board candidate Ref Rodriguez collected $21,000 in campaign donations from employees of his charter school network, Partnerships to Uplift Communities, in his bid to unseat incumbent Bennett Kayser in East Los Angeles’ District 5.

Most striking, a handful of his workers – a janitor, maintenance worker, tutor — are donating at or near the contribution limit, $1,100.

The contributions are a measure of supporters' high hopes to unseat Kayser in favor of Rodriguez, a candidate friendly to charter schools.

Rodriguez, an charter school administrator at Partnerships to Uplift Communities, received most of his financial support from the California Charter School Association Advocates, which received donations from such wealthy donors as former New York Mayor Michael Bloomberg and philanthropist Eli Broad.

Kayser, a former teacher elected as a board member in 2011, collected his largest donations from labor unions, particularly the United Teachers Los Angeles. 

Most of the money working toward Kayser and Rodriguez's reelection are not funneled into their individual campaigns, but to independent expenditure committees which are not subject to the $1,100 contribution limit.

In her first foray into political giving, Luz Maria Lopez, an office worker, donated $1,000 donation to the Rodriguez campaign, twice the amount of Partnerships to Uplift Communities' CEO, Jacqueline Elliot.

“I really believe in Ref. My kids go to PUC schools,” said Lopez, who has been employed by PUC since it opened 15 years ago.  

The employee contributions weren't coerced and will not be reimbursed, Rodriguez said. Many of them can be traced back to a holiday break fundraiser at Rodriguez’s sister’s home in La Puente.

“I know for many of them this is a tremendous sacrifice,” he said. “It’s just been sort of an outpouring of folks belief in me and what we are trying to do for the city.”

Charter school groups major funders

Direct campaign donations from individual contributors, such as Rodriguez’ employees, make up 18 percent of the money spent in the LAUSD’s District 5 school board race. 

The biggest donor is charter school advocacy groups, such as the California Charter School Association Advocates.

Donations have also come from self-described education reform groups that support charter school expansion and firing teachers deemed ineffective, among other issues.

All told, the advocacy groups contributed more than $700,000 to activities in support of Rodriguez and working against Kayser.

On the other side, UTLA funneled $330,000 of members’ contributions to activities supporting Kayser and working against Rodriguez.

While UTLA has turned up its political spending in the board race to stay competitive, it is routinely outspent, said Oraiu Amoni, the union’s political director.

“We never are going to be able to match [reformers] dollar for dollar,” Amoni said. “So our biggest thing is making sure our members are educated, are engaged, are aware — and vote.”

So far, campaigns and committees have spent more than $2 million on the 13 Los Angeles Unified school board candidates, according to filings with the L.A. City Ethics Commission. The contributions have paid for mailing of glossy ads, phone banks, billboards, robocalls and commercials on Spanish-language radio. 

Total contributions are expected to increase in the few days remaining before the primary and swell again in any May runoff. 

Even in major races, aggressive campaigns fueled by growing contributions from special interest groups make it difficult for candidates not affiliated with interest groups to stay competitive.

Limitless independent expenditures are "playing a major role in smaller and local elections,” said Ryan Brinkerhoff, campaign manager for Andrew Thomas, the unaffiliated candidate in the District 5 race.

Thomas, a professor at Walden University, donated $51,000 to his campaign, making him his own biggest contributor. He’s also attracted sizable local support: about 70 percent of his campaign donations come from residents who live in District 5.

Thomas has received no contributions from political action committees or advocacy groups.

Can he win?

“I think so, but it’s getting harder and harder,” Brinkerhoff said. “The results of this election are going to be very telling.”

Outside contributors, local concerns

When public schools were created in the United States, local communities were given control over their governance. Outside money “undermines the relationship between community members and their local public institutions,” according to John Rogers, an education professor at UCLA. 

“It undermines their sense that they own those institutions, and those institutions are theirs to be shaped,” he said.

Without the funds from Broad, Bloomberg and other large donors, Rodriguez’s employees’ contributions would have made up more than 30 percent of his campaign support. Instead, it’s 4 percent.

Kayser has also received support from outside the district, including donations from the American Federation of Teachers and the California Teachers Association.

"The voters have an interest in open and transparent elections in which outside dollars don't have too large an influence," Rogers said.

DC think tank: California online schools group should be investigated

A report released Thursday by a labor group-affiliated Washington think tank is questioning the education provided by an online public school program that says it is in a union fight.

The report by In the Public Interest, a group funded by unions, says the thousands of students enrolled in the California Virtual Academies online public school known as CAVA are receiving a  substandard education by most measures.

"So in every year since CAVA began graduating students, with the exception of 2013, it has produced more dropouts than graduates,” said Shahrzad Habibi, who authored the report.

She said state test score data show that 71 percent of California public schools performed better than the virtual academies.

The report calls on California officials to investigate the online schools’ administration and finances.

California Virtual Academies enrolls about 14,000 kindergarten to 12th grade students through 11 sites, including those in Los Angeles, San Diego, and Fresno. It is run by a national for-profit company called K12 Inc.

In a written statement, California Virtual Academies did not dispute the reported low student performance numbers, but denied other allegations in the study, which it called “inaccurate and deeply flawed.”

“The report relies primarily on misinformation from the California Teachers Association — the union currently engaged in a coordinated and well-funded distortion campaign to unionize the eleven independent California Virtual Academies charter schools.”

In the Public Interest, which supports the work of labor unions, partnered with the American Federation of Teachers last year on a website to track for-profit charter school companies.

Study identifies main culprit behind lithium metal battery failure

A National Science Foundation-funded research has discovered the root cause of why lithium metal batteries fail -- bits of lithium metal deposits break off from the surface of the anode during discharging and are trapped as "dead" or inactive lithium that the battery can no longer access. The discovery challenges the conventional belief that lithium metal batteries fail because of the growth of a layer, called the solid electrolyte interphase (SEI), between the lithium anode and the electrolyte. The researchers made their discovery by developing a technique to measure the amounts of inactive lithium species on the anode -- a first in the field of battery research -- and studying their micro- and nanostructures. The findings could pave the way for bringing rechargeable lithium metal batteries from the lab to the market.

Scientists have recovered the first genetic data from an extinct bird in the Caribbean, thanks to the remarkably preserved bones of a Creighton's caracara in a flooded sinkhole on Great Abaco Island in the Bahamas. Studies of ancient DNA from tropical birds have faced two formidable obstacles. Organic material quickly degrades when exposed to heat, light and oxygen. And birds' lightweight, hollow bones break easily, accelerating the decay of the DNA within. But the dark, oxygen-free depths of a 100-foot blue hole known as Sawmill Sink provided ideal preservation conditions for the bones of Caracara creightoni, a species of large carrion-eating falcon that disappeared soon after humans arrived in the Bahamas about 1,000 years ago. Florida Museum of Natural History researcher Jessica Oswald and her colleagues extracted and sequenced genetic material from the 2,500-year-old C. creightoni femur. Because ancient DNA is often fragmented or missing, the team had modest expectations for what they would find –- maybe one or two genes. But instead, the bone yielded 98.7% of the bird's mitochondrial genome, the DNA most living things inherit from their mothers. The mitochondrial genome showed that C. creightoni is closely related to the two remaining caracara species alive today: the crested caracara and the southern caracara. The three species last shared a common ancestor between 1.2 and 0.4 million years ago. "This project enhanced our understanding of the ecological and evolutionary implications of extinction, forged strong international partnerships, and trained the next generation of researchers," says Jessica Robin, a program director in National Science Foundation's Office of International Science and Engineering, which funded the study.

X-Terra's New Gold Discovery Could Be the Tip of a Large Gold System

Gold has the wind in its sails. Its price in U.S. dollars is up an astounding 62% since late 2015, with a 33% gain in just the past year, outpacing all major assets.

And investors are only just starting to get interested.

The Covid-19 pandemic and its economic impact is a major catalyst. More than $8 trillion in global fiscal stimulus has already been committed to alleviate unemployment and support struggling businesses. But it's almost certainly not enough.

New Brunswick Could Host Large New Gold System

Bona fide new discoveries with district potential are rare. Participating early in one could be a life-changing event.

That's what makes X-Terra Resources Inc. (XTT:TSX.V; XTRRF:OTCMKTS; XTR:FSE) such a compelling investment right now. XTT shares are a Strong BUY, with the potential to double in the next 6–12 months.

Here's my rationale…

Its top two projects are in neighboring Canadian provinces, both among the highest-ranking gold mining jurisdictions globally.

In early March, X-Terra completed its inaugural drill program over the Grog and Northwest Properties in the province of New Brunswick along the McKenzie Fault. It comprised 1,904 meters over 16 holes.

Initial results are in, and they're impressive.

Hole GRG-20-012 identified gold mineralization over a significant width. One interval averaged 0.41 g/t gold over 36 meters, including 0.46 g/t gold over 31 meters and 7.59 g/t gold over 0.6 meters. The company points out that 6 of the remaining holes returned mineralized intervals between 0.1 g/t gold and 0.35 g/t gold.

X-Terra President and CEO Michael Ferreira said, "This is a significant exploration breakthrough, and reinforces our expectations that a large epithermal system is present. While more in-depth geological work, which includes drilling is needed, it remains evident that the 11 holes (1570 metres drilled) only covered a very small fraction of the targeted environment. Reaching a significant mineralized interval this shallow (From 107 metres to 143 metres, in GRG-20-012) is a milestone we were relentlessly pursuing after completing the limited field exploration programs based predominately on roadside trenching. The information obtained in this program will allow the detailed follow up on the Grog Target but also allow the company to refine and generate more high priority targets carrying the same geological characteristics to that of the Grog target. This provides a monumental shift moving forward."

The beauty of this impressive drill hole intercept is its signature, which contains a wide alteration halo associated with sulfidation and quartz veining. Based on the geophysical data, they will be able to track the gold bearing system at depth using an advanced data processing approach combined with their geological knowledge.

The exploration team can now use the signature to formulate similar drill targets elsewhere on the property, with the potential for similar results.

Clearly, X-Terra's diligent, methodical and scientific approach has begun to pay off. Experience combined with a skilled overlay of induced polarization, magnetic surveys, sampling and trenching helped achieve this recent success.

Back in 2017, the company discovered high grade gold occurrences. That was followed up with further work, which delivered extensive anomalies scattered over roughly 30 km along the McKenzie Gulch regional Fault.

Their geologists then engaged a quick exploration cycle over the next 18 months, starting with an orientation geophysics survey, followed by trenching and drilling. They now have an initial model in progress, which involves an extensive magmatic hydrothermal system, and the targets generated so far are pluri-kilometric.

X-Terra is contemplating that it could be onto a brand new regional gold trend.

Such outstanding recent drill intercepts make for an even more exciting outlook. That's because future exploration targets will be chosen with a better understanding of the geological sequence. And that should improve the odds of more successful drill results.

But perhaps the biggest takeaway from hole GRG-20-012 is the suggestion that it demonstrates real potential for a large epithermal system. And that could mean a whole lot of gold lies beneath, something further exploration will answer.

Quebec Offers Huge Promise Near Large Developing Gold Mine

Despite the exciting outlook offered by the Grog area located in New Brunswick, X-Terra is far from being a one-trick pony.

Also bursting with massive untapped potential is the Troilus East Property, located in north-central Quebec.

X-Terra's Troilus East project is immediately adjacent to Troilus Gold Corp.'s former producing gold-copper mine. Even after 15 years of historic production, the Troilus Gold Project currently boasts 4.71 million ounces of gold equivalent in the Indicated category, plus 1.76 million ounces of gold equivalent in the Inferred category.

Early last year, X-Terra announced the completion of a high-resolution magnetic survey on the Troilus-East property. Management continues to advance the project, using the same diligent and methodical scientific approach that has brought success to the Grog discovery. XTT will be using magnetic signatures to perform follow-up work, looking to identify geological contexts with characteristics similar to those of the Troilus gold-copper mineral deposit.

Since tripling its land position, X-Terra has locked up the largest adjacent land claims to Troilus Gold of any public company.

That's exciting, as Troilus Gold is considered by some as the largest—or at least one of the largest—undeveloped gold deposits in North America. And that could well make X-Terra a future target should Troilus Gold or other players look to lock up more of the adjacent land.

People and Projects Offer Massive Potential

As is often the case, people are as important to a junior explorer's success as its properties. As a former professional motorcycle racer, X-Terra President and CEO Michael Ferreira saw the potential of resource exploration to create immense value for shareholders. Now living full-time in the Quebec mining town of Rouyn Noranda, Ferreira has judiciously curated a winning team.

Dr. Michael Byron, Ph.D., P.Geo. and a company director, has thirty years of field work, research and senior management positions across gold, base-metals, diamond and gemstone exploration. He was instrumental in re-discovering Falco Resources' leading asset, the Horne 5 deposit.

That sets up the perfect storm for X-Terra, making it a Strong BUY. With its outstanding initial drill results at the Grog property and the remarkable potential at Troilus East, I can easily see XTT double its market cap in the next 6–12 months, perhaps sooner.

In my view these are the early days of a string of successful exploration results, making XTT.V radically undervalued, for now.

Building a Better Covid-19 Antibody Test

ProMIS is harnessing its unique technology platform to develop a more error-free antibody test.

Testing has been an Achilles heel of the coronavirus pandemic, but ProMIS Neurosciences Inc. (PMN:TSX; ARFXF:OTCQB) has partnered with Dr. Hans Frykman and the BC Neuroimmunology Lab to use its unique technology to create a more accurate antibody test for SARS-CoV-2, the virus that causes Covid-19.

Two main types of tests exist for Covid-19: one that detects the presence of the virus that causes Covid-19, which indicates a person has an active infection, and another that detects antibodies, showing that a person has been exposed to the virus.

The first test that was developed, a test for the presence of the virus, is used mainly to confirm diagnosis of Covid-19 in people who are showing symptoms such as a fever, a dry, persistent cough, difficulty breathing, a sense of restriction in the chest. "They are typical signs of Covid-19, but we would want to know if these are signs of the common flu or a bad cold or Covid-19. We know that Covid can progress really significantly very quickly, especially in individuals with underlying conditions," ProMIS CEO Dr. Elliot Goldstein told Streetwise Reports. "The number of tests is limited, but it's not actually the tests themselves but the reagents and systems you need to run the test that are in short supply."

"Anytime you conduct a test for the virus and get a negative response, the test indicates only that on that day at that time, the person does not have the virus. The person could have had Covid and recovered, or might have had an asymptomatic or very mild case. Or that person could get the virus tomorrow or in three days," Dr. Goldstein explained. "At any point in time the virus test helps indicate the prevalence of the virus—how many people are actually infected—if you test broadly, and at the time you do it, you can determine whether an individual is currently infected or not."

The second type of test, called serological tests or assays, is also known as an antibody test. "When a person is recovering from a viral infection, the immune system makes antibodies—also called immunoglobulins—that are specific to the virus. They neutralize the virus and help clear it out; that's part of the mechanism of why you get better," Dr. Goldstein explained.

One way to see if a person has had Covid is to test for antibodies. "A positive test means you've been exposed to the virus because, in the absence of a vaccine, that's the only way you would have the antibodies. While it's not 100% certain that antibodies neutralize the virus, based on experience with other coronaviruses, it is likely," Dr. Goldstein said. Having the virus neutralized should offer at least some protection against future re-infections.

People who have had positive virus tests know that they have Covid or had Covid and recovered, but many people are asymptomatic or may have had what felt like a light cold, and they want to know if they are at risk, or if they have some protection against the disease. "This is really important for frontline healthcare workers, people working 8-10 hours a day in intensive care or the emergency room with patients known to be very sick with Covid-19; even with protective equipment, they have significant exposure to the virus," Dr. Goldstein explained. "If someone has been through the disease and has natural antibodies, they can't infect someone else. What you want to know on an individual level is am I safe from infection and am I safe for other people."

Generally, antibody testing is a fairly common procedure, Dr. Goldstein explained. When you spin blood in a centrifuge, it separates into three parts: red blood cells, plasma and serum. Serum is where you find antibodies. "ELISA (enzyme-linked immunosorbent assay) is a standard test that looks for antibodies, but it is not specific enough for the Covid-19 virus."

The challenge is there are multiple coronaviruses. "Four different coronaviruses are responsible for the common cold, and then there are others like SARS and MERS. They all have the same sort of halo or corona of protein around the outside of it," Dr. Goldstein said. "They look like the old naval mines used in war. The whole family of coronaviruses look like that. The amino acid sequences of different coronaviruses are not identical but very similar; they share a lot of common structures. There are only really small differences and you can't really pick them up using the usual physical methods."

Studies have shown that up to 90% of individuals in Western countries have been exposed to one or more of the common cold coronaviruses and have antibodies against them. "They look very similar to the coronavirus causing Covid-19. So in Covid-19 antibody tests, the most important thing is it has to be highly specific for the Covid-19 antibodies and doesn't test positive when it identifies a common cold antibody. That is a false positive," said Dr. Goldstein. "It's actually much safer not to have a test that has a lot of false positives because you could base a behavioral decision on faulty information."

Dr. Goldstein cited an example. "If you are testing 1,000 people and there is a 90% prevalence for the cold virus, that means around 900 people have antibodies to the common cold. If the prevalence of the Covid-19 virus is 2%, roughly 20 of the 1,000 would have antibodies to the Covid-19 virus. Let's say the serology test has 95% specificity. That means five times out of 100, it will give a false positive, indicating the presence of Covid-19 antibodies when it is really picking up antibodies against the cold virus. What this means is 5% of 900, or 45 people, will test positive for Covid when they have not had it, and are making decisions based on incorrect information. The consequences of being wrong are dramatic and highlight the need for a very good, high-quality serological test."

How does this relate to Alzheimer's and other neurological diseases that are ProMIS' core competency? "In Alzheimer's, ALS, frontotemporal dementia, Parkinson's disease and other neurological disease, we've been able to use our proprietary, unique technology to identify sites on misfolded proteins that are driving these diseases. Our core technology is the capability to understand what's special about the bad proteins that are causing these diseases and then we can make antibodies highly selective against them. Our technology allows us to identify a region, an epitopes target, which is a series of four to six amino acids where the protein has misfolded. Not only do we know where this target site is located, importantly we also determine the shape (conformation) of this site. Proteins like amyloid and alpha synuclein and TDP 43 misfold and when these proteins misfold they become toxic, they kill neurons, resulting in disease," Dr. Goldstein explained.

ProMIS has transferred that thinking to the virus causing Covid-19. "The corona is composed of the spiky protein. Remember, we want to be able to distinguish between the coronavirus causing the common cold and the coronavirus causing Covid-19," Dr. Goldstein said. "If we can distinguish between the two, we can have an antibody test that's specific for Covid-19. We are looking at a region of the virus called the receptor binding domain, the RBD, that is part of the spike protein and how it attaches to cells. We have a core competency that allows us to identify sites, and not just the location of the sites, but the shape of the sites on complex protein molecules. That allows us then to use that knowledge to create either antibodies or to create serum tests, or even quite frankly, we can use those targets to create vaccines."

Using ProMIS' proprietary technology, the company has been able to "identify a site that we believe is only present on the Covid-19 virus and not on other coronaviruses. We are now initiating the synthesis of several different forms of that site; it's a small area," Dr. Goldstein stated. "That would then transfer to Dr. Hans Frykman's lab at University of British Columbia, a world-class serology lab. Then we will see if the targets we've identified are specific and selective antibodies against Covid-19."

When you test the serum of an individual, if they've been exposed to the virus and have the antibodies, "those antibodies should bind selectively and specifically to the target. So if the antibodies from the patient's serum are binding to the target site, we know it's a Covid-19 virus because that site is only visible in that shape on the Covid-19 virus and not the others. For the validation of our test, only in patients known to have had Covid-19 should we see binding of antibodies against Covid-19 to our target. The second validation is based on testing in serum from subjects known to have never been exposed to Covid-19 virus—such subjects have antibodies only from cold or other coronaviruses, and therefore the antibody test should be negative; there should be no binding. So we should only see binding in serum from a patient known to have recovered from COVID-19, and we should not see binding in serum from an individual known not to have been exposed to COVID-19," Dr. Goldstein explained.

"Our technology basically allows us to zero in with sniper-like precision on the structure of a protein and understand it, not only the structure overall but the shape of the regions on that protein and then that allows us to identify what is specific to that protein, in this case the spiky protein on the virus causing COVID-19," said Dr. Goldstein.

Texas Oil & Gas Firm Achieves EBITDA, EPS Beats in Q1/20

A recap of Parsley Energy's Q1/20 performance and projections for this year and next are given in a Raymond James report.

In a May 5 research note, analyst John Freeman reported that Raymond James increased its target price on Parsley Energy, Inc. (PE:NYSE) after it posted its Q1/20 numbers.

Raymond James' new target price on Parsley is $12 per share, up from $11. The Texas-based energy company's stock is trading now at about $9.38 per share.

Freeman reviewed and commented on Parsley's Q1/20 results. The company "delivered modest EBITDA and earnings per share beats relative to the Street" due to oil pricing," Freeman pointed out.

Production was relatively in line at 126,600,000 barrels of oil per day (126.6 MMbbl/d), which was 1% higher than consensus' forecast but 1% below Raymond James' estimate. Total production was 1% above the Street's projection but 3% below Raymond James' forecast.

"The performance on the quarter was encouraging, however, the highlight from earnings was the significant reduction in 2020 capex (down from about $1 billion to less than $700 million)," Freeman commented.

Capex, "a welcome surprise," Freeman wrote, came in 5% and 7% lower than the investment bank and the Street's estimates, respectively. Opex was 3% under Raymond James' projection

Moreover, Parsley's related maintenance capital needs were greatly below expectations as well, indicating that Parsley made capital efficiency gains during the period.

"We were pleasantly surprised that Parsley is able to maintain in line Q4/20 oil volumes (about 115 MMbbl/d) on a capital program that's about $300 million/30% below the Street," added Freeman.

Looking forward, Raymond James modeled a base case, or stable scenario for Parsley, that implies a West Texas Intermediate oil price of about $30 a barrel and Parsley having four to five rigs and one to two crews operating. In that scenario, Parsley would produce about 117 MMbbl/d in 2020 and 115 MMbbl/d in 2021.

Capex would amount to about $678 million in 2020, dropping to $598 million in 2021.

Free cash flow would be about $300 million in 2020, which coincides with Parsley's guidance of $300M plus, and increasing to $370 million in 2021.
