We've all been saying it: These are unprecedented times. The impacts of the COVID-19 pandemic are incredibly wide-ranging and affect all facets of life. One that is hitting the scientific community very hard is the cancellation of meetings, large and small. While we are well-versed in connecting with colleagues and collaborators across a variety of online platforms, these do not replace the immensely gratifying aspects of attending meetings in person: the pleasure of catching up with old friends and making new ones, the insights gained from having real-time conversations with others working on the same topic but with different expertise and perspectives, and the stimulating new scientific ideas we carry home. We have all been feeling the disappointment as we learn that one meeting after another is forced to cancel, from the vibrant ASBMB annual meeting to summer conferences of all types.Another loss from the appropriate but painful decision to cancel the ASBMB annual meeting was the chance to hear from our Herbert Tabor Early Career Investigator Awardees, who represent the best science published in JBC in the preceding year. This year, the competition was particularly fierce. We hope and anticipate that we will be able to hear from the winners at next year's ASBMB annual meeting. But in the meantime, we want to raise a toast to Wenchao Zhao, Yue Yang, Manisha Dagar, Febin Varghese, and Ayumi Nagashima-Kasahara as our 2020 winners. Repression of sphingosine kinase (SK)-interacting protein (SKIP) in acute myeloid leukemia diminishes SK activity and its re-expression restores SK function [Molecular Bases of Disease]

Previous studies have shown that sphingosine kinase interacting protein (SKIP) inhibits sphingosine kinase (SK) function in fibroblasts. SK phosphorylates sphingosine producing the potent signaling molecule sphingosine-1-phosphate (S1P). SKIP gene (SPHKAP) expression is silenced by hypermethylation of its promoter in acute myeloid leukemia (AML). However, why SKIP activity is silenced in primary AML cells is unclear. Here, we investigated the consequences of SKIP down-regulation in AML primary cells and the effects of SKIP re-expression in leukemic cell lines. Using targeted ultra-HPLC-tandem MS (UPLC-MS/MS), we measured sphingolipids (including S1P and ceramides) in AML and control cells. Primary AML cells had significantly lower SK activity and intracellular S1P concentrations than control cells, and SKIP-transfected leukemia cell lines exhibited increased SK activity. These findings show that SKIP re-expression enhances SK activity in leukemia cells. Furthermore, other bioactive sphingolipids such as ceramide were also down-regulated in primary AML cells. Of note, SKIP re-expression in leukemia cells increased ceramide levels 2-fold, inactivated the key signaling protein extracellular signal-regulated kinase, and increased apoptosis following serum deprivation or chemotherapy. These results indicate that SKIP down-regulation in AML reduces SK activity and ceramide levels, an effect that ultimately inhibits apoptosis in leukemia cells. The findings of our study contrast with previous results indicating that SKIP inhibits SK function in fibroblasts and therefore challenge the notion that SKIP always inhibits SK activity.

Mechanistic insights explain the transforming potential of the T507K substitution in the protein-tyrosine phosphatase SHP2 [Signal Transduction]

The protein-tyrosine phosphatase SHP2 is an allosteric enzyme critical for cellular events downstream of growth factor receptors. Mutations in the SHP2 gene have been linked to many different types of human diseases, including developmental disorders, leukemia, and solid tumors. Unlike most SHP2-activating mutations, the T507K substitution in SHP2 is unique in that it exhibits oncogenic Ras-like transforming activity. However, the biochemical basis of how the SHP2/T507K variant elicits transformation remains unclear. By combining kinetic and biophysical methods, X-ray crystallography, and molecular modeling, as well as using cell biology approaches, here we uncovered that the T507K substitution alters both SHP2 substrate specificity and its allosteric regulatory mechanism. We found that although SHP2/T507K exists in the closed, autoinhibited conformation similar to the WT enzyme, the interactions between its N-SH2 and protein-tyrosine phosphatase domains are weakened such that SHP2/T507K possesses a higher affinity for the scaffolding protein Grb2-associated binding protein 1 (Gab1). We also discovered that the T507K substitution alters the structure of the SHP2 active site, resulting in a change in SHP2 substrate preference for Sprouty1, a known negative regulator of Ras signaling and a potential tumor suppressor. Our results suggest that SHP2/T507K's shift in substrate specificity coupled with its preferential association of SHP2/T507K with Gab1 enable the mutant SHP2 to more efficiently dephosphorylate Sprouty1 at pTyr-53. This dephosphorylation hyperactivates Ras signaling, which is likely responsible for SHP2/T507K's Ras-like transforming activity.

Prominins control ciliary length throughout the animal kingdom: New lessons from human prominin-1 and zebrafish prominin-3 [Cell Biology]

Prominins (proms) are transmembrane glycoproteins conserved throughout the animal kingdom. They are associated with plasma membrane protrusions, such as primary cilia, as well as extracellular vesicles derived thereof. Primary cilia host numerous signaling pathways affected in diseases known as ciliopathies. Human PROM1 (CD133) is detected in both somatic and cancer stem cells and is also expressed in terminally differentiated epithelial and photoreceptor cells. Genetic mutations in the PROM1 gene result in retinal degeneration by impairing the proper formation of the outer segment of photoreceptors, a modified cilium. Here, we investigated the impact of proms on two distinct examples of ciliogenesis. First, we demonstrate that the overexpression of a dominant-negative mutant variant of human PROM1 (i.e. mutation Y819F/Y828F) significantly decreases ciliary length in Madin–Darby canine kidney cells. These results contrast strongly to the previously observed enhancing effect of WT PROM1 on ciliary length. Mechanistically, the mutation impeded the interaction of PROM1 with ADP-ribosylation factor–like protein 13B, a key regulator of ciliary length. Second, we observed that in vivo knockdown of prom3 in zebrafish alters the number and length of monocilia in the Kupffer's vesicle, resulting in molecular and anatomical defects in the left-right asymmetry. These distinct loss-of-function approaches in two biological systems reveal that prom proteins are critical for the integrity and function of cilia. Our data provide new insights into ciliogenesis and might be of particular interest for investigations of the etiologies of ciliopathies.

The mRNA levels of heat shock factor 1 are regulated by thermogenic signals via the cAMP-dependent transcription factor ATF3 [Metabolism]

Heat shock factor 1 (HSF1) regulates cellular adaptation to challenges such as heat shock and oxidative and proteotoxic stresses. We have recently reported a previously unappreciated role for HSF1 in the regulation of energy metabolism in fat tissues; however, whether HSF1 is differentially expressed in adipose depots and how its levels are regulated in fat tissues remain unclear. Here, we show that HSF1 levels are higher in brown and subcutaneous fat tissues than in those in the visceral depot and that HSF1 is more abundant in differentiated, thermogenic adipocytes. Gene expression experiments indicated that HSF1 is transcriptionally regulated in fat by agents that modulate cAMP levels, by cold exposure, and by pharmacological stimulation of β-adrenergic signaling. An in silico promoter analysis helped identify a putative response element for activating transcription factor 3 (ATF3) at −258 to −250 base pairs from the HSF1 transcriptional start site, and electrophoretic mobility shift and ChIP assays confirmed ATF3 binding to this sequence. Furthermore, functional assays disclosed that ATF3 is necessary and sufficient for HSF1 regulation. Detailed gene expression analysis revealed that ATF3 is one of the most highly induced ATFs in thermogenic tissues of mice exposed to cold temperatures or treated with the β-adrenergic receptor agonist CL316,243 and that its expression is induced by modulators of cAMP levels in isolated adipocytes. To the best of our knowledge, our results show for the first time that HSF1 is transcriptionally controlled by ATF3 in response to classic stimuli that promote heat generation in thermogenic tissues.

5-Ethynyl-2'-deoxycytidine and 5-ethynyl-2'-deoxyuridine are differentially incorporated in cells infected with HSV-1, HCMV, and KSHV viruses [Microbiology]

Nucleoside analogues are a valuable experimental tool. Incorporation of these molecules into newly synthesized DNA (i.e. pulse-labeling) is used to monitor cell proliferation or to isolate nascent DNA. Some of the most common nucleoside analogues used for pulse-labeling of DNA in cells are the deoxypyrimidine analogues 5-ethynyl-2'-deoxyuridine (EdU) and 5-ethynyl-2'-deoxycytidine (EdC). Click chemistry enables conjugation of an azide molecule tagged with a fluorescent dye or biotin to the alkyne of the analog, which can then be used to detect incorporation of EdU and EdC into DNA. The use of EdC is often recommended because of the potential cytotoxicity associated with EdU during longer incubations. Here, by comparing the relative incorporation efficiencies of EdU and EdC during short 30-min pulses, we demonstrate significantly lower incorporation of EdC than of EdU in noninfected human fibroblast cells or in cells infected with either human cytomegalovirus or Kaposi's sarcoma-associated herpesvirus. Interestingly, cells infected with herpes simplex virus type-1 (HSV-1) incorporated EdC and EdU at similar levels during short pulses. Of note, exogenous expression of HSV-1 thymidine kinase increased the incorporation efficiency of EdC. These results highlight the limitations when using substituted pyrimidine analogues in pulse-labeling and suggest that EdU is the preferable nucleoside analogue for short pulse-labeling experiments, resulting in increased recovery and sensitivity for downstream applications. This is an important discovery that may help to better characterize the biochemical properties of different nucleoside analogues with a given kinase, ultimately leading to significant differences in labeling efficiency of nascent DNA.

Study shows need for new focus in anti-vaping efforts for older teens and young adults

(Michigan Medicine - University of Michigan) They know it's addictive. They know it's linked to dangerous lung diseases. And they know it delivers more nicotine than the cigarettes it's supposed to replace. But the social aspects of vaping drives young people to use Juul and other e-cigarettes, according to nearly two-thirds of teens and young adults in a new study. Less than 5% say the availability of fruity flavors drives use of e-cigarettes by members of their generation, and only 10% say addiction does.

Shoreline clean-up conducted

Various government departments, together with volunteers today conducted a shoreline clean-up along the remote rocky beach in Kung Pui Wan, Tap Mun to remove two tonnes of refuse.

 

The Environmental Protection Department, the Food & Environmental Hygiene Department (FEHD) and the Marine Department participated in the operation.

     

The Inter-departmental Working Group on Marine Environmental Management said the beach, facing the windy and wavy sea, is not easily accessible by working vessels and the rough terrain connecting the rocky beach also increases the difficulty of routine cleaning work.

 

FEHD cleaners along with the volunteers packed the refuse and delivered it on foot to a nearby pier for temporary storage. FEHD staff then took the refuse to the Marine Department's collection vessel in batches for onward delivery to a rubbish collection point for centralised handling.

     

To minimise the risk of spreading COVID-19, the operation was carried out in groups of no more than four participants each. They maintained an appropriate distance from each other and paid heed to personal protection, including wearing masks.

     

The working group thanked the volunteers for taking part and called on the public to keep the countryside and shoreline clean during outings.

HK airport’s fresh approach

In March, Hong Kong International Airport (HKIA) became the first in the world to be certified as an International Air Transport Association (IATA) Center of Excellence for Perishable Logistics (CEIV Fresh) Partner Airport.

 

Two of the airport’s cargo terminal operators, Hong Kong Air Cargo Terminals Limited (HACTL) and Cathay Pacific Services Limited (CPSL) have achieved IATA CEIV Fresh Certification, while Cathay Pacific recently became the world’s first airline to be certified.

 

IATA created the Centre of Excellence for Independent Validators (CEIV) to help organisations involved in the air cargo supply chain achieve handling excellence.

 

The certification underscores the speed, consistency and efficiency of all HKIA stakeholders when it comes to the treatment and transportation of perishable products.

 

It also helps to ensure food safety, and reduce food wastage and loss along the supply chain.

 

Quality benchmark

Hong Kong Air Cargo Terminals Limited Chief Executive Wilson Kwong said the main value of the accreditation is that it provides a universally recognised standard.

 

“It gives assurance to the various stakeholders that we have, and we are adhering to the highest possible standards.”

 

Both accredited cargo terminal operators HACTL and CPSL have set up perishable cargo handling areas with truck docks next to the apron.

 

As soon as the perishable goods are unloaded from the aircraft, they are immediately transported to the handling area.

 

The clearance and sampling checks have been streamlined to minimise the time taken to unload the goods. There are cold rooms set at tailored temperatures for various kinds of perishable cargo.

 

Their staff training and handling process have also been assessed and certified for their compliance with the global industry standards.

 

Cathay Pacific Services Limited Chief Executive Officer Jenny Lam noted that one of the challenges in meeting the certification standard is training its 2,000 employees in handling and storing perishable goods.

 

“They first have to identify what types of perishables they are, such as are they flowers, are they fruits, are they meat, so that they know how to segregate the storage.

 

“Secondly, they gain knowledge about certain fruits that cannot be put close to each other such as bananas and apples.”

 

Growth opportunities

In 2018, the airport handled about 380,000 tonnes of perishable cargo - a rise of 12% on 2017, with perishables accounting for about 8% of total air cargo volume.

 

Airport Authority Commercial Executive Director Cissy Chan explained that temperature-controlled air freight is one of the fastest growing segments in the air cargo market.

 

She believes with the new certification, the airport is well-placed to capture the increasing growth opportunities in the market for fresh and perishable goods.

 

“With the opening of the Hong Kong-Zhuhai-Macao Bridge, the transport time between the airport and the Greater Bay Area, notably the western part of the Pearl River Delta, will be further cut down.

 

“So this will put us in an even better position to become the transhipment hub for these perishables for the entire bay area.”

 

Extra backing

To support the growing demand for perishables, the Airport Authority has provided additional cool dollies, so there are now more than 30 for use by the whole airport community. It is also building apron shelters to protect the perishables from ambient weather elements.

 

Cathay Pacific Cargo Service Delivery General Manager Frosti Lau said they would work closely with the authority and its cargo terminal to see what new investments are required to handle the increasing demand.

Redressing HK with eco-fashion

The modern day phenomenon of fast fashion has completely changed consumer and retailer behaviour.

 

The latest trends fresh off the catwalk are now available off the rack in record time and for a fraction of the price. But these garments are usually made of cheap, low quality materials that fall apart after a few wears and end up in the landfill.

 

Fast fashion’s impact on the environment is huge. This is where environmental non-governmental organisation Redress plays a role.

 

Drawing board

Launched in 2007, Redress works to reduce waste in the fashion industry. It promotes sustainability through organising fashion shows, seminars and exhibitions in Hong Kong and abroad.

 

In 2011, the EcoChic Design Award was born. Relaunched as the Redress Design Award in 2018, it is open to emerging fashion designers and design students all over the world. The award aims to inspire designers to create mainstream clothing with minimal textile waste.

 

Redress Circular Fashion Programme Director Kay Liu explained the motivation behind setting up the award came from discovering that 80% of a product’s environmental impact is determined during the design stage.

 

“That means designers have a lot of influence on how their product is going to impact the environment, so we really want to spread the message for fashion designers to understand the power they have and to positively impact the industry,” Ms Liu added.

 

The competition works to educate emerging fashion designers around the world about sustainable design theories and techniques to drive growth towards a circular fashion system.

 

In the space of a decade, the award has evolved from a local competition to the largest global platform for talented fashion game changers to transform the fashion industry worldwide.

 

Fashion muse

Keith Chan is one of those young design talents. His collection, inspired by Hong Kong’s signature neon signs, won him the Hong Kong Best Prize for 2019.

 

Mr Chan said his fellow competitors inspired him to try out different sustainable design techniques. A designer from Spain used natural plant dyes to colour her garments and changed his perspective on the industry’s dyeing methods.

 

“This is not only about design, you also need to consider our society, really think about the materials you use and how to make your outfit better,” Mr Chan noted.

 

Fashion design students like Jasmine Leung were also inspired through joining the competition.

 

Each competition cycle takes participants on an educational journey lasting several theory and design-packed months to educate designers about fashion’s negative environmental impacts.

 

By attending seminars and completing various tasks for her collection, Ms Leung gained a better understanding of eco-fashion.

 

“The award gave me an opportunity to acquire practical fashion knowledge beyond the classroom. I have learnt about circular design strategy, like applying the concept of zero waste, recycling and upcycling.”

 

Major milestone

The Redress Design Award marks its 10th cycle this year. Many of its alumni have gone on to establish more than 40 sustainable brands globally, with some already receiving international recognition.

 

A new addition to this year’s contest is the Redress All-Star design competition which is open to alumni from past cycles to mark the milestone birthday. Applications for the Redress Design Award 2020 officially opened on January 8.

 

To celebrate the 10th cycle, a retrospective exhibition is also being held at The Mills until January 19, where visitors can get a close-up look at all the winners’ creations.

UBC researchers establish new timeline for ancient magnetic field on Mars

As businesses slowly reopen, experts warn that social distancing may need to be dialed back up: ‘It’s not an on-off switch’ * Coronavirus – latest US updates * Coronavirus – latest global updatesSome California businesses on Friday began opening their doors for business – at least partially.As states and counties across the nation contend with pressure to lift the stay-at-home measures that have destroyed local economies, California is taking an especially cautious approach, walking a fine line between political and economic pressure to reopen and the public health imperative to stop the spread of disease.Public health experts told the Guardian that while no US state was equipped with enough coronavirus testing and surveillance to feel fully confident reopening, California’s slow, piecemeal recovery plan – though far from perfect – seemed like the least risky option. The planSeven weeks after the governor, Gavin Newsom, ordered his 40 million constituents to shelter in place and all non-essential businesses to close, California on Friday entered phase two of its grand reopening plan.Some retail stores, including bookshops, florists, music stores, clothing and sporting goods retailers, can reopen if they organize curbside pickup. Some manufacturing and logistics in the retail supply chain can restart as well, as long as they follow safety and hygiene protocols. And local authorities are allowed to ease regulations further than the state guidelines if they meet certain testing and sanitation requirements.Phase three of the plan – potentially months away – could see salons, gyms, movie theaters and in-person church services resume. Phase four would end all restrictions. The timingFriday’s reopenings come as California has avoided the surge of infections states like New York have seen. And although California has seen more than 61,000 cases and 2,500 deaths, its hospitals have not been overwhelmed.Last week, state officials reported the first week-over-week decline in Covid-19 deaths.The new guidelines also follow small but sustained protests across the state to demand a relaxation of regulations to revive the state’s crippled economy, and some rural counties have partially reopened in defiance of the lockdown measures. The caveatsHowever, California still hasn’t seen the two weeks of declining cases that the White House suggested as a criterion for easing restrictions and that several European countries have used as a benchmark.The state also lacks the robust testing and tracking systems that countries such as Germany and South Korea have used.The state has ramped up its ability to administer and process tests, although for now, its rate of 29,414 tests a day is below the figure required by some analyses.Authorities are working to put a robust contact tracing effort in place to make sure those who test positive get the care they need and are able to isolate themselves until they recover. Although some counties and communities have spearheaded community-wide testing and tracing programs, overall, the state isn’t at the point where its system is as widespread or efficient as a country like Germany’s.Experts say California should also have a system in place to make sure vulnerable, unhoused populations have access to shelter and medical care – to prevent infection flare-ups in homeless shelters and encampments. Progress on those measures heavily varies county by county.And ideally, there would be a treatment or a vaccine before reopening, said Dr Richard Jackson, a professor emeritus at the UCLA Fielding School of Public Health and the former head of the California department of public health. While we await a cure, Jackson cautioned, “no one should feel completely safe as we remove restrictions.” The trade-offsCalifornia’s reopening strategy stands in sharp contrast to the approach of states like Georgia, which suddenly allowed gyms, barber shops, hair salons, tattoo parlors and bowling alleys to welcome customers last week.“What certain places have done, where they’ve just thrown open the doors and said, ‘OK, we don’t have to keep our distance any more,’ is a colossal mistake,” Jackson said. Reopening businesses that put lots of people into close contact and speed the spread of disease will reverse the success of shelter-in-place rules, he noted, and overwhelm hospitals as cases surge. “Doing it very cautiously and carefully does make sense at this point in time,” he said.“I get that governors have to balance the public health goals with the economic goals,” said Dr Robert Tsai, surgeon and health policy researcher at Brigham and Women’s hospital in Boston and the Harvard TH Chan School of Public Health. “But this stage of the pandemic is really all about trade-offs,” he noted. The weeks aheadIn the coming weeks, state and local leaders will have to watch closely and prepare to dial the distancing back up if the number of cases surges, said Tsai.“Social distancing isn’t an on-off switch. What it needs to be is a dial, which can be turned up or down depending on what the data show on the ground in terms of how the Covid-19 epidemic is progressing.“Reopening is going to be a very complicated process, and it should be complicated,” he added. “Because this is about making sure that people don’t end up in the hospital or dying.”That California’s plan allows for counties to maintain stricter distancing guidelines or ease up measures could be both a strength and a liability.The flexibility has allowed hotspots like the Bay Area and Los Angeles to take a more cautious approach, but it has also already caused confusion. In San Diego, where curbside shopping has already begun, business owners were unsure what, if anything, would change on Friday. In Bakersfield, restaurants allowed patrons to dine in on Monday and Tuesday, in defiance of the state’s guidelines.A hodgepodge reopening could cause surges in cases; Californians who travel between more lax and more strict counties could spread infections. Moreover, a rush to reopen fast in some areas could be counterproductive to economic recovery, said Alessandro Rebucci, an economist at the Johns Hopkins Carey School of Business.“If you reopen when the pandemic is still out there, people and businesses will not just go back to normal,” Rebucci noted. Based on research from China, it seems clear that fear of contracting the illness will keep businesses owners and patrons home until they feel it’s safe enough, he said.
