Near infrared fluorogen and fluorescent activating proteins for in vivo imaging and live-cell biosensing

Tissue slices and whole organisms offer substantial challenges to fluorescence imaging. Autofluorescence and absorption via intrinsic chromophores, such as flavins, melanin, and hemoglobins, confound and degrade output from all fluorescent tags. An “optical window,” farther red than most autofluorescence sources and in a region of low hemoglobin and water absorbance, lies between 650 and 900 nm. This valley of relative optical clarity is an attractive target for fluorescence-based studies within tissues, intact organs, and living organisms. Novel fluorescent tags were developed herein, based upon a genetically targeted fluorogen activating protein and cognate fluorogenic dye that yields emission with a peak at 733 nm exclusively when complexed as a “fluoromodule”. This tool improves substantially over previously described far-red/NIR fluorescent proteins in terms of brightness, wavelength, and flexibility by leveraging the flexibility of synthetic chemistry to produce novel chromophores.

The invention relates to a method for producing a protective layer on a surface of a plant, to a protective layer for a surface of a plant, to a plant coated with said protective layer, to a composition for carrying out the method and for producing the protective layer, and to uses of said composition. According to the invention, a method is proposed, wherein at least one sol gel having nano-scale particles is formed by the hydrolysis of at least one precursor in water and at least one nano-scale layer of the sol gel is applied onto the surface of the plant. The protective layer according to the invention comprises a nano-scale SiO2 layer, and the composition according to the invention comprises at least one SiO2-producing substance.

Weed control in joints of concrete block and other paving stone

The present weed reducing material reduces the necessary weed control work by making joint filling sand a hostile substrate for plant and fungus growth by using an environmentally acceptable slow release mineral additive. The additive, having similar grain size distribution, mechanical and rheological properties, forms an integral part of the joint filling sand. The specific gravity of the additive is lower than quartz sand imparting a tendency for the additive to migrate towards the top part of the joint. The additive leads to pore waters rich in sodium and at high pH, both qualities being maintained over long periods. The mixture of additive and sand is handled and applied using conventional laying techniques and equipment, observing usual precautions for mortar or cement mixes with sand. In the environment, reaction with CO2 in the air or in the soil porosity inactivates the causticity, yielding harmless carbonate salts.

A method for promoting the health of a plant comprises administering malic acid to the plant or the soil in an amount effective to recruit plant growth promoting rhizobacteria (PGPR) to the plant. Administration of malic acid promotes biofilm formation of PGPR on the plant's roots, thereby restricting entry of a foliar pathogen through stomatal pores present in the leaves. Another method for promoting the health of a plant comprises administering acetoin to the plant or the soil in an amount effective to increase pathogen resistance in aerial parts of the plant.

Compositions and methods, including novel homogeneous microparticulate suspensions, are described for treating natural surfaces that contain bacterial biofilm, including unexpected synergy or enhancing effects between bismuth-thiol (BT) compounds and certain antibiotics, to provide formulations including antiseptic formulations. Previously unpredicted antibacterial properties and anti-biofilm properties of disclosed BT compounds and BT compound-plus-antibiotic combinations are also described, including preferential efficacies of certain such compositions for treating certain gram-positive bacterial infections, and distinct preferential efficacies of certain such compositions for treating certain gram-negative bacterial infections.

Molecular flux rates through critical pathways measured by stable isotope labeling in vivo, as biomarkers of drug action and disease activity

The methods described herein enable the evaluation of compounds on subjects to assess their therapeutic efficacy or toxic effects. The target of analysis is the underlying biochemical process or processes (i.e., metabolic process) thought to be involved in disease pathogenesis. Molecular flux rates within the one or more biochemical processes serve as biomarkers and are quantitated and compared with the molecular flux rates (i.e., biomarker) from control subjects (i.e., subjects not exposed to the compounds). Any change in the biomarker in the subject relative to the biomarker in the control subject provides information to evaluate therapeutic efficacy of an administered drug or a toxic effect and to develop the compound further if desired. In one aspect of the invention, stable isotope-labeled substrate molecules are administered to a subject and the label is incorporated into targeted molecules in a manner that reveals molecular flux rates through metabolic pathways of interest.

Polymeric blends having improved flame retardance properties and good ductility at low temperatures are disclosed. The invention is directed to methods for determining clonotypes and clonotype profiles in assays for analyzing immune repertoires by high throughput nucleic acid sequencing of somatically recombined immune molecules. In one aspect, the invention comprises generating a clonotype profile from an individual by generating sequence reads from a sample of recombined immune molecules; forming from the sequence reads a sequence tree representing candidate clonotypes each having a frequency; coalescing with a highest frequency candidate clonotype any lesser frequency candidate clonotypes whenever such lesser frequency is below a predetermined value and whenever a sequence difference therebetween is below a predetermined value to form a clonotype. After such coalescence, the candidate clonotypes is removed from the sequence tree and the process is repeated. This approach permits rapid and efficient differentiation of candidate clonotypes with genuine sequence differences from those with experimental or measurement errors, such as sequencing errors.

Cardiac pacing methods for an implantable single chamber pacing system, establish an offset rate for pacing at a predetermined decrement from either a baseline rate (i.e. dictated by a rate response sensor), or an intrinsic rate. Pacing maintains the offset rate until x of y successive events are paced events, at which time the offset rate is switched to the baseline rate for pacing over a predetermined period of time. Some methods establish a preference rate, between the offset and baseline rates, wherein an additional criterion, for switching from the offset rate to the baseline rate, is established with respect to the preference rate.

A method is disclosed for preparing a soft tissue site, and augmenting the soft tissue site, such as the breast(s), scar, depression, or other defect, of a subject through use of devices that exert a distractive force on the breast(s) and grafting of autologous fat tissue such as domes with sealing rims for surrounding each of the soft tissue site and a regulated pump. The method for preparing the soft tissue site, and enhancing fat graft results, entails application of the distracting force to the targeted soft tissue site at least intermittently for some period of time and preferably several weeks prior to the graft procedure. Disclosed is a mold hanger for supporting a bottle mold in a blow molding station, the mold hanger comprising a piston and piston sleeve fully contained within the mold hanger configured to push a moveable insert into the mold. Also disclosed is a method of retrofitting an original rotatable blow molding module having multiple existing blow molding stations, each existing mold hanger defining an existing outer envelope. The disclosed method may include providing an improved mold hanger substantially contained within the respective existing outer envelope and including low-profile drive mechanisms configured opposably to drive moveable inserts into the mold. Further disclosed is a method of manufacturing a blow molded bottle with a deep pinch grip, the method including providing within a mold hanger a drive mechanism to drive a moveable insert into the mold. A bottle made by such methods is also disclosed.

Hot surface igniter adaptive control method

An adaptive gaseous fuel ignition control method for use in consumer and commercial appliances that reduces stress on and increases the life of a hot surface igniter without resulting in a failure to ignite condition is provided. The method provides a preheating period, a full temperature period, and a trial for ignition period. The preheating period gradually increases the power applied to reduce the stress resulting therefrom. Once a gas valve has been commanded open, the controller monitors the time for ignition of the gaseous fuel. If the time is longer than a threshold, either the applied power or the period of time during which the power is applied is increased to shorten the time. If, however, the ignition period is shorter than the threshold, either the power applied or the period of time during which the power is applied before commanding the gas valve open is lowered or shortened.

A remotely actuated pilot gas valve includes safe lighting and complete shutoff capabilities in the event that the flame that is heating a thermocouple is extinguished. The invention provides for a heater system that utilizes such a pilot gas valve as well as a method whereby the pilot gas valve used in such a system can be remotely and electronically actuated when required. In addition, the gallium phthalocyanine crystal is used in the photosensitive layer of the electrophotographic photosensitive member.
